Objective:To discover the effects and mechanism of action of Guizhi fuling pill with flavor guilie on the morphological structure and levels of harmful substances of the dorsal root lymph nodes of rats suffering from painful diabetic peripheral neuropathy.Provide new research sight and direction for diabetes treatment of Chinese medicine.Materials and methods:104 healthy SPF grade male SD rats weighing approximately200 g to 220 g were used.Adapted diet for one week,12 rats were randomly selected as a blank control group and the remaining rats were modelled for diabetes.Following a 12-hour fast,fasted body weight was determined in the moulded rats,followed by an intraperitoneal injection of a 2% streptozotocin solution at a dose of 50mg/kg,which induced the rats to become a model of diabetes.The venous venous blood sugar was measured three days later.Rats with a glucose concentration greater than 16.7 mmol/L were included in the follow-up study.Fed with ordinary foods,lasting two model group(P<0.01),and there was no significant difference between the Yikang group and the model group(P>0.05).3.Compared to the normal group,the rats in the model group experienced slow weight growth compared to each administered group,with significant weight loss at 4 weeks of administration(P<0.01);The body weight of rats treated at 8 weeks with medium and high doses of Jiawei Guizhi Poria Pills increased somewhat in relation to the model group(P<0.01).There was no statistically significant difference between the yikai group and the model group.4.Compared with the normal group,the nerve conduction velocity of the model group and the other drug groups decreased significantly after 4 weeks of treatment(P<0.01),and the nerve conduction velocity of the high-dose treated group was higher than that of the model group after 8 weeks of treatment(P<0.01).5.Before administration,compared with the normal group,the 50% paw withdrawal threshold of the model group and each treatment group was significantly decreased(P<0.01),and there was no significant difference between the model group and each treatment group(P>0.05).After 4 and 8 weeks of treatment,the 50% paw withdrawal threshold of the model group was significantly lower than that of the normal group(P<0.01).Compared with the model group,the 50% paw withdrawal threshold of the middle and high dose treatment groups of Jiawei Guizhi Fuling pill was increased(P<0.01).6.Compared with the normal group,the licking time of the model group and the treatment groups was significantly shortened(P<0.01).There was no significant difference in licking time between the model group and the treatment groups(P>0.05).After 4 and 8 weeks of treatment,the licking time of the model group was significantly shorter than that of the normal group(P<0.01).Compared with the model group,the licking time of rats in the Jiawei Guizhi Fuling pill low,medium and high dose treatment groups was significantly increased(P<0.01).7.Compared to the normal group,the expression of 5-HT and HA in serum of model group was significantly increased(P<0.01);Compared to the model group,the expression of 5-HT and HA in serum of rats in treatment groups was significantly decreased(P<0.01).8.Compared with the normal group,the interstitial Spaces of the dorsal root lymph nodes of L4-6 in the model group rats were enlarged and disorganized,with some of the lymph nodes separated from the satellite glial cells.However,the morphology of the treatment groups was better than that of the model group.Conclusion: Guizhi Fu Ling pill can improve the mental state and behavior of PDPN rats with painful diabetic peripheral neuropathy,reduce blood glucose,increase body weight,increase nerve conduction velocity,alleviate the sensitivity of PDPN rats to pain,repair the damaged nerves,and reduce the expression level of pain-causing substances 5-HT and HA in the serum of rats.In order to relieve PDPN pain mechanism.
