The Human Genomic Project is a coordinated major international research effort with the ultimate goal, by the year 2005, of having sequenced the entire human genome, identified the estimated 10 000 human genes and researched the genome of model organisms. The first and important step for reaching the goal is to create genetic map and develop physical map, including the construction of YAC, P1, PAC, BAC and cosmid clone contigs based on their human genome library. The second generation of genetic map containing 5 264 markers known as microsatellites or short tandem repeat polymorphisms marks the completion of genetic map. The first generation of physical map consisting of average 0.9 Mb YAC contig and the second -generation of YAC contigs of single chromosome such as 3, 12. 21, 22 and Y have been constructed, but it can not well satisfy the needs of genome analysis in details. On other hand, the cosmid contig of chromosome 16 have been generated, which especially attracts our attention, because it starts to describe the assembly of maps at even higher resolution and cosmid clones are the appropriate vector for sequencing and cloning genes. Therefore, a massive task for next phase of Human Genomic Project is the construction of maps of the genome at cosmid level. An important part of the generation of accurate physical maps is to use the same set of markers to identify and correct any inconsistency.
