Activation of glutamine synthetase gene (glnA) plays a pivotal role inbacteria in response of nitrogen starvation. Here we show that GlnR of A.mediterranei U32, is a direct activator of glnA gene, but enriched innitrogen excess condition. It was also showed that A. mediterranei U32GlnR is involved in the global nitrate-effect on the rifamycin SVproduction. Using a low-copy vector, A.mediterranei U32 GlnR wasintroduced into its phylogenetically related organism, Streptomycescoelicolor, and we found it widely participated in host strain's secondarymetabolism, while totally deletion or increment the copy of its nativeGlnR did not display such pleiotropic effects. Our study suggests thatGlnR is able to either promote or inhibit its target gene's transcription, aworking model for GlnR is proposed.
