Treatise on febrile disease is one of the classical works of the traditional Chinese medicine (TCM), as prescriptions in it were strictly organized with good curative effect in consequence,they are called"classical prescriptions"and are used widely in generations. However,because of the diversity of disease,the succession of era and the system of weights and measures,doctors of later generations often used them by adjustment on medication’s dosage or number,which depend on era’s characteristics and personal experience. The situation has confused the system of original dosage, megadose and slight dose are coexisted. Gegen Qinlian Tang,Huangqin Tang and Baitouweng Tang are all commonly-used classical prescriptions,dosage control of the three also face the trouble,moreover,as the three prescriptions are mutually-used frequently,how to rebalane the new prescriptions by managing dosage is another problem to solve. This problem is ubiquitous existed nowadays and tightly-connected with clinical ptractice. Proper dosage is essential for treatment,no matter Chinese medicine or western medicine. Compare to the accurate guideline of western medicine,the way of controlling dosage of TCM is more empirical and vague,some principles are not clear enough for doctors or students of TCM to handle,because of the lack of specific standard. Confront with this situation,this dissertation focus on the medical cases and literature that related to Ge Gen Qin Lian Tang,Huang Qin Tang and Bai Tou Weng Tang,through the statistics and analysis on the dosage of the three prescription in clinical usage,and related influencing factors as well,attempt to seek out methods and rules which concerning with the medication dosage control,and can be more visualized,objective and convenient,for the clinical reference.Objective:(1)Statistically analyse the dosage of Gegen Qinlian Tang,Huangqin Tang and Baitouweng Tang in medical cases from Ming dynasty to nowadays,including dosage of single classical prescriptions itselves, dosage and proportion of every medication of the three prescriptions, with the comparison of the whole prescriptions’dosage. Based on this, analyse and quantify the specific influence on medication’s dosage from the prescriptions’structure and effect, medication’s interaction,patient’s age and gender, time and course of disease,symtoms and signs,then work out the methods,summarize the rules.(2)Take the three prescriptions as a whole,attempt to excavate certain disciplinary pattern and method for dosage control and composition,then build a simulation model for dosage forecasting of the three prescriptions.(3)Distinguish prescriptions by medication’s dosage and reveal the rules in it, analyse the similarities and differences of certain medication’s dosage among prescriptions.(4)Seek out frequently-used additive medication by statistics,test the interaction between them and the one from the three prescriptions,probe the format that how they participate into the three prescriptions’ patterns.(5)Build a simulation model to forcast the content of related medicinal composition on account of the prescription dosage of Gegen,Huangqin,Huanglian and Gancao,compare the variation of every medicinal composition’s content under two different dosage compatibility proportion,discuss the way for dosage control and reasonableness test of composition from the angle of relationship between the prescription dose and decoction dose.(6)Collect data information by the exercise and assessment of many machine learning arithmetic and statistical approach, in order to supply reference for correlational research.Contents:(1)Medical cases that were recorded from Ming dynasty to June 2014,take Gegen Qinlian Tang,Huangqin Tang or Baitouweng Tang as main prescription,or contain at least three medications of the aim prescriptions,and in the mean time, all total medications of the whole prescription in the case would not more than eight.(2)Literature that established from 1949 to June 2014,associated with admeasurement of the medicinal composition’s content of Gegen Qinlian Tang.Methods:(1)Data collection and entry:according to the inclusion and exclusion criteria,search and select medical cases and literature in "TCM dictionary",medical cases books and CNKI,WanFang,VIP,CBM,remove repetitive ones,then enter them into two databases that were built in advance.(2)Data pre-processing:including the standardization of terms,such as diagnosis,pattern type,symptom,medication name,etc;transforming words,such as patient’s age,gender, time and course of disease,symtom and sign,into number in two ways,one depends on symptom or sign’s existence,another depends on symptom or sign’s degree;unity the unit of medication and medicinal composition.(3)Data analysis:on K-Miner system,choose suitable analytical method according to contents’charaterisitics.Results:(1)Prescriptions dosage:dosage range of Gegen Qinlian Tang,Huangqin Tang and Baitouweng Tang are all broad,and the dosage proportion also vary a lot from the original,moreover.additive medications are used commonly.The roles of medications in Gegen Qinlian Tang are basically the same,but the dosage gap among medications decline,especially Gegen and Huanglian’s.In Huangqin Tang, dosage proportion of Shaoyao increase sharply.sometimes even catch Huanglian’s level.In Baitouweng Tang,Baitouweng and Qinpi’s dosage rise,while Huanglian’s gets a Significantly decline.(2)Medications dosage:①dosage of the one that used to eliminate pathogenic factors is more extensive then the one that used to strengthen body resistance, dosage of adult and children all correspond with this characteristic basically.Besides.maximum dosage of adult is higher than children’s,but the minimum dosage of adult is nearly at the same level,or even lower than children’s,which reflect that the dosage range of adult is more broad than children’s. Details of main section including:build a simulation model by the decision tree classification algorithm,which is used for classifying the three prescriptions by the composed medications dosage,the classification rules are:if Gegen Qinlian Tang is the main prescription,then Gegen’s dosage should more than 3.0g,if take Baitouweng as additive medication,then its dosage should less than 6.0g,or cannot have Huangbai in the same time when Baitouweng’s dosage is over 6.0g;if Baitouweng Tang is the main prescription,then Baitouweng’s dosage should over 0, if take Gegen and Huangqin as additive medications,then Gegen’s dosage should less than 3.0g and Huangqin’s should less than 12.5g,if Qinpi is in the prescription,then there is on limit for Huangqin’s dosage;②Huangqin and Gancao’s dosage in Gegen Qinlian Tang are different from Huangqin Tang,Huanglian’s dosage in Gegen Qinlian Tang and Baitouweng Tang are nearly the same;③through the correlation analysis on medications’ interaction,gain three functional expression,including:Gegen’s dosage=0.95785103×Huangqin’s dosage+1.52477879, Baitouweng’s dosage=1.08781865×Huangbai’s dosage+3.54663593, Baitouweng’s dosage=1.06093496×Qinpi’s dosage+2.51797048, Huangbai’s dosage=0.5444367×Qinpi’s dosage +0.75104051,besides,get several correlation coefficients that represent the interaction between medications;④by attribute importance ratings and correlation analysis algorithm,quantify the specific influence on the dosage from symtoms and signs, patient’s age and gender, time and course of disease.(3)Dosage control related to composition pattern:①have 10 patterns in all by clustering algorithm analysis on the medications of the three prescriptions and additive medications that frequently-used;② by support vector regression,build and test a stimulation model for forcasting medications’s dosage by patient’s information including symtoms and signs,patient’s age and gender, time and course of disease,the result of test is relatively positive.(4)Prescription dose-decoction dose:use artificial neural network and generalized linear regression,build a stimulation model for forcasting main medicinal composition’s content of Gegen,Huangqin,Huanglian and Gancao,by its prescription dose,then test it by two dosage proportion of Gegen Qinlian Tang(5:3:3:2 and 8:3:3:2),the result is reasonable with supporting the rationality of the 8:3:3:2 proportion,therefore,this procedure may become a potential way to check a prescription’s rationality.Conclusion:Results above successively clarify the clinical dosage use of Ge Gen Qin Lian Tang,Huang Qin Tang and Bai Tou Weng Tang,excavate and discuss related methods and rules for clinical medication dosage control,and quantify some of it,thus provide more objective and convenient reference for the clinical medication dosage control of the three prescriptions. On the other hand, according to the TCM theory,some parts of the results are not exactly reasonable, which are mainly due to the data quantity and quality,method and processing of data preprocessing, insufficient knowledge reserves of researcher,etc. Towards these factors,propose and discuss the corresponding scheme for improvement,then envision the development trend and key point in the research of TCM medication dosage control.
