Huangqin-Tang decoction was one of the most widely used traditional Chinese medicines, composed of Radix Scutellariae,Radix Paeoniae Rubra,Glycyrriza and Fructus Zizyphi Jujubae. Recent reports in the scientific literature have indicated that Huangqin-Tang has the pharmacodynamics effect such as resist bacteria and inflammation, adjust immune system, ease pain, protect the liver, lower the index of aminotransferase, anti-cancer and virus, lower the physical temperature, and calm down the emotion. The original and relative prescription can be commonly used in curing intestines disease such as diarrhea, acute enteritis and gastritis, ameba diarrhea, etc. It can also be used in internal medicine, gynecology, dermatology, etc.There were many compounds in Huangqin-Tang, In conventional studies, a few reference compounds had been used to control the quality or research the pharmacokinetic of Huangqin-Tang. However, these studies were not able to reflect the decoction's whole efficacy completely, because the complicated constituents may work synergistically, which was not combinated with each herb simply. In the fact that the species in blood and the possible metabolites which demonstrate to originate from the prescription are responsible for the observed efficacy, Based on the progress of studies on serum Pharmacochemistry for traditional Chinese medicine (TCM) and hypothesis that only the constituents migrating to blood after orally TCM may have the probability to be the effective constituents, serum pharmacyochemistry was strategically applied to study the effective compounds in rats'plasma after oral administration of Huangqin-Tang. Concering the comparative study of Huangqin-Tang in the first components of the entry point as the breakthrough point, the transfer of blood-forming and active ingredients in the metabolic process and the effective components for the preparations has been investigated. through HPLC fingerprints, serum Pharmacochemistry and pharmacokinetics, the effect components of Huangqin-Tang can been initially clarified. the improvement of the scientific quality standards and the compatibility theory has laid the foundation of interpretation.1. A practical chromatographic method was established on rats'plasma after oral administration of Huangqin-Tang. The chromatographic separation was carried out on Diamonsil(?)C18 (4.6mmx250mm,5μm). Precipitation proteide with three times methanol was chosen as a sample preparation method, for it can removed a lot of endogenous impurities and it can also extracted most of the target compounds.2. An analytical approach of ascribing constituents migrating to blood after oral administration of Huangqin-Tang was established. Learning from fingerprint strategy, rats'plasma samples after oral administration of Huangqin-Tang and individual herb were compared in different conditions. According to the retention time determined by HPLC in the condition of Scutellaria baicalensis, eight compounds in rats'plasma were ascribed, including five of them came from herb, three of them were metabolites. And according to the retention time determined by HPLC in the condition of Red Paeony Root, nine compounds in rats'plasma were ascribed, including three of them came from herb, six of them were metabolites. And according to the retention time determined by HPLC in the condition of LiquoricRoot, six compounds in rats'plasma were ascribed, including three of them came from herb, three of them were metabolites.3.By identifying the constituents migrating to blood after oral administration of Huangqin-Tang by HPLC-MS. We can obtain much valuable information. Due to the advantages of high sensitivity and specificity, LC-MS(ESI/APCI)was applied to identify and determine the compounds'structure in plasma after oral administration of Huangqin-Tang. On the basis of the retention time, UVmax and m/z in positive or negative ion mode, five compounds including Baicalin, Wogonoside,Paeoniflorin,Glycyrrhizic acid and Glycyrrhetinic acid were identified or tentatively elucidated comparing with literature.Which would to be useful to us.To sum up, the modern high performance liquid chromatography (HPLC) coupled with mass spectrum (MS) identification was successfully used on the study of the serum pharmacochemistry of Huangqin-Tang. With the fingerprint strategy, five compounds were identified and a efficient and significative process was developed, which provided a new method for analyzing the effective compounds in TCM and It would have broad prospects.
