Effects Of Xuefu Zhuyu Decoction On Pharmacokinetics Of Pueraria Lobata In Normal And Blood Stasis Rats

Objectives:With normal rats and blood stasis rats as experimental animals, to study the effect of peoniflorin pharmacokinetics with platycodon root and root of bidentate achyranthes in Xuefu Zhuyu decoction; and to set up the method of HPLC with the extract of Xuefu Zhuyu decoction and the measurement of density in peoniflorin from the serum of rats; In order to explore the effect of platycodon root and root of bidentate achyranthes as messenger drug.Methods:With the opposite method of HPLC:chromatographic column is Diamonsil C18column(250*4.6mm,5μm); the temperature of column is30℃; the mobile phase is acetonitril water(16:84); the current speed is1.0ml/min; the examination wavelength is230nm, the objects of examination is peoniflorin and riboflavin is the internal standard. Male and female rats with180-230g weight were divided into normal group and blood stasis group. Started the blood stasis group on I.V2%dextran with T500saline,2ml/kg in order to make blood stasis model. Without food for12h but with water freely, both of groups rats stomachs were perfused with the whole Xuefu Zhuyu decoction and decoction without platycodon root and root of bidentate achyranthes, every dose for6rats with11380μg/kg. After5min,10min,15min,20min,30min,45min,1h,2h and4h, the normal rats’blood were taken by orbital venous plexus; the blood stasis model rats stomachs were perfused after30min and then after another10min,15min,20min,30min,45min,1h,1.5h,2.5h and4h, their blood were taken by orbital venous plexus. When the measurement of density of blood had been finished, to analyze pharmacokinetics with DSA2.0, and the pharmacokinetics parameter was counted by SPSS13.0.Results:After perfused by whole Xuefu Zhuyu decoction, normal rats’AUC(0-t) is0.348mg/L*h, t1/2is0.671h, Tmax is0.276h, CL is68.423L/h/kg, V is41.303L/kg, and Cmax is0.362mg/L. After perfused by Xuefu Zhuyu decoction without platycodon root and root of bidentate achyranthes, normal rats’AUC(0-t) is0.135mg/L*h, t1/2is1.175h, Tmax is0.317h, CL is84.068L/h/kg, V is113.053L/kg and Cmax is0.104mg/L. After perfused by the whole Xuefu Zhuyu decoction, blood stasis rats’AUC(0-t) is 0.286mg/L*h, t1/2is0.583h, Tmax is0.5h, CL is50.109L/h/kg, V is39.075L/kg and Cmax is0.317mg/L. After perfused by Xuefu Zhuyu decoction without platycodon root and root of bidentate achyranthes, blood stasis rats’ AUC(0-t) is0.326mg/L*h, t1/2is0.711h, Tmax is0.542h, CL is48.761L/h/kg, V is35.785L/kg and Cmax is0.256mg/L.Conclusion:The effect of platycodon root and root of bidentate achyranthes in Xuefu Zhuyu decoction can increase the speed in normal rats intaking peoniflorin, and add up the density peoniflorin in serum of normal rats, but there is no obvious effect on peoniflorin pharmacokinetics in blood stasis rats.
