Doctoral postgraduatd : Pan Xi-long Advisor : Qiu HelmingThe ternu Causes and Pathogenesis of Acute Leukemia (AL) have been studied and drawn some Certain by traditional Chinese medicine (TCM) .AL belongs to consumption , hemorrhagic diseases and seasonal frbrile diseases in TCM .The Pathogenesis is deficiency in origin and excess in Superficiality. Deficiency in origin includes deficiency of qi and blood. Excess in superficiality includes stagnation of qi, blood stasis and accumulation of noxious matter. Traditional Chinese Medicine believes that pathogensis of malignant tumor in blood stem after chemotherapy is hyperactivity of heat-toxicity , disorder of Qi and blood .unbalance of spleen and stomach, deficiency of liver and kidney Yin, so,there is an emergency stage of heat-toxicity, mixing deficiency, phlegm, blood stasis .In the guide of TCM, there is two patent herbs Qingdu y/MQDY) consisting of herbs of clearing away heat and toxic materials .. activating blood circulation to dissipate blood stasis % eliminating phlegm and removing stasis .. cooling blood and stop bleeding Yangzheng p/a/?(YZP) consisting of herbs of supplementing qi and nourishing blood> replenishing yin and essence. Generally speaking, QDY has functions of resisting tumor , and increasing immune function , YZP hasfunctions of improving myelosuppression, protecting liver and kidney, QDY and YZP treated different stages of AL together or singly according to differentiation of sydromes .Post clinical studies showed complete remissionccr (CR) rates were high, the time . reaching CR were short, the total effects were good in QDY and YZP groups . Post eaperimental studies revealed model mousy ,survival stages were prolonged in experimental groups ,QDY and YZP could improve immune function, raise the activaties of interlukin 2(IL-2) > IL-6 and tumor necrosis facter alpha (TNF-a), raise the mRNA expression of the IL-2, IL-6,TNF-d . To explore the role of QDY and YZP in Apoptosis in Leukemia, We built up leukemic model of mice by inoculate leukemia cells,it divided into Seven qroups leukemia model group:normal mice group;QDY group;YZP group; chemical drug group;QDY add chemical drug group, YZP add chemical drug group.The apoptosis were observed with terminal dutp-mediated nick end labeling (TUNEL),The outcomes were that more apoptotic cells were found in QDY group, YZP group, chemical group,QDY add chemical drug group and YZP add chemical drug group (P<0.05),QDY group had more apoptotic cells than YZP, QDY add chemical drug group had the most apoptotic cells, The apoptotic index(AI) in every experimental groups was obvious larger than that in model group (P<0. 01), It showed That QDY and YZP all could impel Leukemic cell apoptosis, and could raise the effect of chemistry ,and QDY was better than YZP, Liver index and spleen index were also observed , it is found that, Liver index and spleen index in QDY group, YZP group, chemical group , QDY add chemical drug group and YZP add chemicaldrug group decreased significantly than that in model group (P<0. 05),(it is showed that QDY and YZP had effect of anti-leukemia ,and could increase the effect of chemistry.The protein expression of Bcl-2 - P53 .. Fas, were detected with immunohistochemistry and special staining , the outcome were that the protein expression of Bcl-2 in 17212 model mice was increased , the protein expression of Bcl-2 in QDY group , YZP group , chemical group , QDY add chemical group and YZP add chemical group decreased markedly (P<0.05), The protein expression of Fas in QDY group, YZP group, chemical group ,QDY add chemical group and YZP add chemical group increased obviously (P<0. 05), and QDY was better than YZP . The protein expression of P53 in every experimental group increased for some extent , but the difference has no statistic significance (P>0. 05), The serum Fas (sFas) was examined with enzyme linked immunosorbent assays (ELISA) , the outcome was .comparing to model group , sFas in QDY group decreased obviously (P<0.05), comparing...
