Objective: Varicocele is the primate cause leading to male infertility. Testicular pathological changes bring out sperm maturity disturbance, sperm quality decreasing, but they are short of acknowleged quantu mark. According to Makler score, we have studied seminiferous tubule's pathological alteration in rats with varicocele, and made a scientific appraisal to breeding sperm function, which benefit to male infertility's diagnosis and treatment. Methods: 30 male S-D rats were selected at random, they were classified as two groups:20, as varicocele group; 10,as sham operation group. Varicocele mode was created by Saypol's method.Three months after operation,rat's spermatic vein was observed at the state of anesthesia,operating microscope measure vein's diameter.Bilateral veins had manifested certain varicose, particularly the left, the left diameter0.63±0.03mm, the right 0.36±0.04mm, they had significant differences (P<0.05). In controlled group, bilateral veins' diameter had no alteration, they all were 0.30±0.03mm, testicular tissue were stained and fixed routely. Under microscope, 10 seminiferous tubule break faces were observed in each sample. We assayed seminiferous tubule bore(D), peri-membrane depth(M), dissepiment germ cell's layer number(P) and germ cell maturity(S). Every item's full mark was 5,4 items accounted for 20. The average score(TMS) also could be calculated.Results: In left testis, VG's seminiferous tubule bore(D) were diminuted significantly than SoG's, they were 97.65±11.66um and 143.56±10.72um respectively. Peri-membranes(M) become thicker(4.70±0.28um vs 2.04± 0.25um), cell layer number (P) decreased (2.23±0.58 vs 5.36±0.59), germ cell's maturity(S) became disturbant (3.43±0.67vs 4.77±0.47), IMS was lower than SoG's (10.15±1.14 vs 13.10±1.20), they had significant differences.The left testicular breeding sperm function was compared with the right one's in VG, the left D,M,P,S were 97.65±11.66, 4.70±0.28, 2.23±0.58 , 3.43±0.67 ;the right were 113.72±9.94 , 3.47±0.22 ,3.14±0.59 , 4.04± 0.45,respectively,they all had significant differences.The left testicular breeding sperm function was also compared with the right one's in SoG, the left D,M,P,S were 143.56±10.72, 2.04±0.25 , 5.36± 0.59, 4.77±0.47 ; the right were 144.90±10.42, 2.01±0.23 , 5.13 ±0.54 , 4.79±0.43, they had no significant differences(P>0.05)Conclusions:1. Testicular pathological quantitative study had manifested that varicocele could lead to breeding sperm functional lesion, which brought out seminiferous tubule bore become decreasing, peri-membrane turned thicker, cell layer number decreased, sperm maturity were disturbed, TMS decreased.2. Makler score was applied to appraise breeding sperm function,which was objective and reliable,it deserved expending.
