PART I Cell viability and HSP-70 synthetic activity treated by hyperthermia and chemotherapeutic agent in both normoxia and hypoxia.Section I Cell viability and HSP-70 synthetic activity treated by hyperthermia and chemotherapeutic agent in normoxiaPurpose The aim of the present study was to evaluate the cell viability and compare heat shock protein HSP-70 synthetic activity treated by hyperthermia and chemotherapy agent in normoxia. Methods The cell viability was evaluated by MTT method after hyperthermia at 37,39,41,43℃ for 30 minutes or combined with 50mg/ml MMC or 2% lipiodol in human hepatocellular carcinoma cell line HCCLM3 as well as rabbit carcinoma VX2 cells and human umbilical vein endothelial cells. Detection of heat shock protein HSP-70 in HCCLM3 cells two hours later under different temperature at 37,39,41,43℃ for 30 minutes by western blot. HSP-70 was also examined at 1, 2, 4, 6, 8, 12 hours after heated by 41℃ for 30 minutes in HCCLM3 cells. Result: The cell viability measured by OD value treated by hyperthermia was decreased by ascending temperature in HCCLM3 cells as well as VX2 and human umbilical vein endothelial cells. Human umbilical vein endothelial cell viability was significant lower compared with HCCLM3 and VX2 cells. HSP-70 protein expression was detected after heated at 39, 41, 43℃ for 30 minutes. HSP-70 protein synthetic activity was up to 6 hours after hyperthermia at 41℃ for 30 minutes. Conclusion: The cell viability measured by OD value treated by hyperthermia was decreased by ascending temperature in HCCLM3 cells as well as VX2 and human umbilical vein endothelial cells.HSP-70 synthetic activity was initiated and last for 6 hours after hyperthermia stress, HSP-70 may play a substantial role in cell response to hyperthermia stress in normoxia.Key words: cell viability; MTT; OD value; HSP-70; Western blot; normoxia...
