Intraperitoneal hyperthermia chemoperfusion(IHCP) was first used in clinic by Spratt in 1980.It is useful in treatment for stomach cancer which invase or metastses to the peritoneum by clinic trial report, but the mechanisms of hyperthermic chemotherapy for different type gastric cancer and the role of Fas and bcl-2 induced by hyperthermic chemotherpy should be under further observation and research.The propose of this paper is to do some experimental research in vivo and in vitro on these questions and to understand more about the mechanisms of hyperthermia chemotherapy on different type gastric cancer cell,providing theoretical basis to improve the effectiveness of hyperthermia chemotherapy in clinic.Experimental study in vitro一, The growth inhibit effect of hyperthermic chemotherapy on SGC7901 and MKN45object: Compare the difference of growth inhibit effect of hyperthermic chemotherapy on different differentiation type gastric cancer cell linesmethod: We used two-differentiation stage of gastric cancer cell lines (SGC7901 and MKN45) under hyperthermia, chemotherapy and hyperthermia chemotherapy treatment respectively in vitro.Acid phosphatase assay was used to compare the growth inhibition effect of the three kind of therapies on the same stomach cell lines. Also we compared the growth inhibition effect of one kind therapy on two differentiation type gastric cancer cell lines.results: hyperthermic chemotherapy has stronger growth inhibit effect on middle SGC7901 than the poor differentiated gastric cancer cell line MKN45二, The effect of hyperthermic chemotherapy treatment on cell morphology and cell cycle for SGC7901 and MKN45object: To observate the characteristics of cell mophology and changes of cell cycle in two differentiated gastric cancer cell lines following hyperthermic chemotherapymethod: We used flow cytometer to compared the influence of hyperthermic chemotherapy on the tumor cell cycle and observe the cell morphotogy on optic microscope and electronic microscoperesults: The G1 , G2 stage cells are remarkably increased and S stage cells decreased after hyperthermic chemotherapy in SGC7901, a lot of vacuole was noticed in SGC7901 under electronic microscope as well as optic microscope. Cell cycle changing were not significant after hyperthermic chemotherapy in MKN45.a less vacuole was found in MKN45 under electronic microscope as well as optic microscope.三, The apoptosis rate of SGC7901 and MKN45 induced by hyperthermic chemotherapyobject: to investigate the effect of hyperthermia chemotherapy on apoptosis of diffferent type gastric cancer cell SGC7901 and MKN45. Method: two human gastric cancer cell line SGC7901 and MKN45 were cultured in vitro and underwent hyperthermia chemotherapy. the apoptosis rate measured by TUNEL Results: the apoptosis rate of SGC7901 after hyperthermic chemotherapy 48h later and 72h was 42.38±0.12%and 50.17±0.41% respectively while 32.89±0.23% and 40.37±0.39% respectively of MKN45.the P value was significantly different in statistics study. 四, The variance of expression level of apoptosis regulate gene Fas in hyperthermic chemotherapyobject: To compare the expression levels of Fas both SGC7901 and MKN45 in IHCPmethod: Hyperthermia, chemotherapy and hyperthermic chemotherapy treatment were applied on two differentiation type gastric cancer cell lines in vitro. We used CD95PE to label the two cancer cell lines. Flow cytometer was applied to analysis the expression level of Fas generesults: the expression level of Fas gene was significantly higher in SGC7901 after chemotherapy and IHCP, especially by IHCP.
