Objectives: Heat shock protein (HSP) is a ubiquitous highly conserved polypeptide protein family. It consists of several subfamilies, including HSP27, HSP60, HSP70 and HSP90 according to their molecular weight. Their main function is to sustain the stability of proteins essential for cellular viability, which is considered as functions on "housekeeping" and cell protection and important role of regulation. Recently, the relationship of HSP with diseases, especially tumors, has been attracted increasingly more attention. Studys on roles of HSP in apoptosis of tumor cells and anti-tumor immunity and prognosis of patients with caicinomas have been hot topics for researchers on oncology and immunology. Among members of HSP, HSP27 and HSP70 have been found to correlate closely with prognosis of cancer. Researches have demonstrated that over-expressions of HSP27 and HSP70 in gastric carcinoma, breast cancer and prostate carcinoma correlated inversely with depth of invasion, tumor differentiation and lymph node metastasis. Patients with high expression of HSP27 and HSP70 tended to have a poor prognosis. However, there were only a few reports on expression of HSP27 and HSP70 in esophageal squamous cell carcinoma (ESCC). The prognostic significance of expression of HSP27 and HSP70 in ESCC is not completely understood. The potential mechanisms responsible for their prognostic effect on ESCC are not clear. Esophageal carcinoma (EC) is one of common malignances in China. It is the fourth cause of cancer deaths just following gastric cancer, liver cancer, and lung cancer. The morbidity of EC is of geographical feature. Hebei province is a high incidence area of EC. The main pathological type of EC in China is squamous cell carcinoma. During past decades, the diagnosis and treatment of ESCC have been greatly improved and the postoperative death rate after esophagectomy has markedly decreased. But, the prognosis of patients with ESSC is still relatively poor, especially those with in advanced stages. Long-term survival rate is quite disappointing. Therefore, how to accuratly predict and obviously improve prognosis of ESCC and increase the long-term survival rate of ESCC patients have been one of medical focuses in the past decades. Up to now, works on the expression of HSP27 and HSP70 and their significance in esophageal squamous cell carcinoma are few, while the results were controversial. To further explore the prognostic significance of expressions of HSP27 and HSP70 in ESCC, 101 cases of ESCC with detail clinical pathological and follow-up data were included in this study. The cases were divided into two groups that were matched in all pathological and clinical features but post-esophagectomy survival time according to follow-up data. The following studies were carried out:1 The expression and clinical significance of heat shock proteins 27 and 70 in esophageal squamous cell carcinomas with different post-esophagectomy survival peroid.2 The clinical significance of NK cell and DC infiltration in esophageal squamous cell carcinomas with different post-esophagectomy survival peroid and their relationship with expression of HSP27 and HSP70.3 The expression and prognostic significance of the expression of survivin and caspase-3 in esophageal squamous cell carcinomas with different post-esophagectomy survival peroid and their relationship with expression of HSP27 and HSP70.4 The expression of HSP27 mRNA in esophageal squamous carcinoma, mucosa adjacent to cancer and normal esophageal mucosa.Methods: 1 The expression and clinical significance of heat shock proteins 27 and 70 in esophageal squamous cell carcinomas with different post-esophagectomy survival peroid The expression and clinical significance of HSP 27 and HSP70 in 101 cases of ESCC were studied with S-P immunohistochemical method. Among the 101 cases, 38 cases were classified into long-term survival group (≥5 years) and 63 cases into short-term survival group (≤1 year) according to following-up data.
