They were Aged/Ethanol group, aged control group, Young/Ethanol group and young control group. Ethanol groups was perfused 50% alcohol(v/v) on the dose of 12ml/kg twice per day instead of water in control groups. Nine rats died in Aged/Ethanol group and one died in Young/Ethanol group. All rats were dissected after 7 days and total RNA were isolated from their liver and cerebral cortex samples. Then PAI-1 mRNA was amplificated by RT-PCR and compared to-actinmRNA.Results Mortality increased significantly in Aged/Ethanol group than Young/Ethanol group(P<0.05). The expression of PAI-1 mRNA significantly increased in Ethanol groups compared to control groups(P<0.01) in rat's liver. The same thing happened when Aged/Ethanol group compared to Young/Ethanol group(P<0.01) and aged control group compared to young control group(P<0.05). The rate of positive expression of PAI-1 mRNA in rat's cerebral cortex significantly increased in Aged/Ethanol group compared to aged control group(P<0.01). The same thing happened when Aged/Ethanol group compared to Young/Ethanol group(P<0.01) , Young/Ethanol group compared to young control group(P<0.05) and aged control group compared to young control group(P<0.05).Conclusion Both age and alcohol can induce the over-expression of PAI-1 mRNA in rat's liver and cerebral cortex. The effect of alcohol is enhanced in aged rats.
