Peptic Ulcer (PU) is a commonly-encountered chronic recurrent disease.With the application of H2 receptor antagonist (H2-RA) and proton pumpinhibitor (PPI) in recent years, the ulceration got faster short-term healing.However,65%-80% of the patients got relapse within one year afterdiscontinuity of the drugs, and almost 100% relapse within 2 years;Helicobacter(Hp) eradication therapy could reduce the annual recurrence rateof Hp-related ulcer to less than 3%, but the recurrence rate turned out to behigh because of such problems as cost, side effects and the compliance ofpatients. In addition, part of the ulcerations had nothing to do with Hpinfection, therefore it's become urgent to prevent recurrence of peptic ulcer. Research materials showed that the fully-recovered ulcer had a lowrecurrence rate, especially the concept of "quality of ulcerative healing"(QOUH) on the International Digestion Conference in Hawaii by Tarnawski atthe end of 1990, which changed the people's understanding about ulcerrecurrence. Since then more and more scholars started to do research onQOUH. And thus it's become the key point for anti-recurrence to improveQOUH. Clinical study indicated that Chinese herbs could achieve the similarrecovery rate with H2-RA in treating PU, but without toxic-side effects.Furthermore, it has been noticed that the recovered ulcer cases by Chineseherbs had a low recurrence rate. Unfortunately there is not enough research onimproving QOUH. Based on the traditional knowledge on PU by TCM and modern researchachievement, we produced Wei-xi Kang, the pure Chinese herbal preparationthrough the method to supplement spleen, clear heat, regulate stomach andpromote descending and got good clinical effect. In order to clarify theanti-ulceration effect of the formula and study its treatment mechanism, we 13英 文 摘 要duplicated the model rats of gastric ulcer (GU) with the injection of acetic acidand intended to study its effect on histology maturation degree of regeneratedmucosa through ulcer index (UI), maturation degree of the tissue andultrastructure; to study its effect on the maturation degree of regeneratedmucosa function in GU rats through the effect on GMBF, NO, ET-1, EGF,TGF- α , EGFR, PGE2 and hexosamine; to study itsinhibition-killing-interference on Hp through the bacteria-inhibitionexperiment on Hp; to study the anti-reinjury and anti-recurrence mechanism ofWei-xi Kang through observing the long-term effect on reinjury by ethanoland ulcer healing. Method Part I Duplication of acetic acid GU model and function of EGF during automatic repair Experiment 1 Duplication of acetic acid GU model and photoscope observation on GU mucosa Twenty-one cleaning male Wistar rats weighed from 220 to 250g wererandomly divided into three groups (100% acetic acid 10μl, 50μl andnormal group, 7 rats in each group). And the ulcer model were duplicated byinjection considerable dosage of glacial acetic acid through the rats' anteriorwall of gland and stomach close to pylorus and the optimal dosage wasdetermined through general and tissue photoscope observation. Experiment II Study on Expression of EGF, TGF-α and EGFR in Self-Healing of GU Rats Forty-two cleaning male Wistar rats weighed from 220 to 250g wererandomly divided into two groups, one is normal group (6 rats) and the otheris 6 observation groups (36 rats).
