It has been regarded in TCM that five viscera are center of life activity of human body, the deficiency of five viscera is the fundamental reason for aging. Gradual deficiency of essence in kidney is closely related to brain aging , the kidney stores essence from five-Zang viscera and six-Fu viscera while spleen dominates tranformation and transportation and it is in charge of sending up the clear, which is called the origin of the acquired constitution and the source for generating Qi and blood. Five viscera all depend on its physiological functions, as a result, the sea of medullae must be nourished by the acquired constitution. After middle age, descending of Qi is more than ascending. Failure of ascending of clear Qi results in malnutrition of brain, mental weariness and amnesia. Hence, replenishing spleen-stomach is one of the most important methods to postpone aging of brain and body. Meanwhile, deficient spleen is apt to generate dampness and accumulate phlegm, accumulation of phlem causing stagnation of Qi or deficiency of spleen causing insufficiency of Qi could in turn lead to blood stasis. Phlegm, turbidity and stasis are important factors affecting aging. Both the methods ofreplenishing and discharging should be taken according to the deficiency -excess theory.ObjectiveThe subject is based on the theory that spleen-stomach is the origin of the acquired constitution, with replenishing spleen-stomach as a treatment, and it carries out two methods which are replenishing spleen Qi, replenishing spleen Qi added with dispelling phlegm and stsis. It aims at observing the function and mechanism of the two methods for preventing and treating brain aging, and also having two methods compared, which offer the theoretical support and experimental basis for senescense and brain aging due to spleen deficiency.Method1. select 96 adult 2.5-month mice of kunming species, divided randomly into normal control group, model control group , herb group A and herb group B (24 for each) . Every group except normal control group has been subcutaneously administrated with a big doze of D-gal through injection at posterior neck for 6 weeks continuously in order to make subacute senescense and brain aging animal model. Herb group A were given replenishing spleen Qi decoction and herb group B were given replenishing spleen Qi added with dispelling phlegm and stasis decoction.2. Observe change of capability of learning and memory of mice with aging brain by the means of Morris water maze at the 37th day of experiment,after 6 days of observation, take samples and assay the following indexes:(1) Superoxide dismutase (SOD) in serum, heart and liver was assayed by subnitric acid salt method. Methylenedioxyamphetamine (MDA) in heart and liver tissue was assayed by TEA method. Liposcin (LPF) hi heart and liver was assayed by fluorescence method.(2) Expression level of nicotinic acetylcholine receptor(nACh-R) and glutamate N-methyl-D-aspartate receptor(NMDA-Rl) of cerebral cortex and hippocampus are assayed by immunohistochemical SABC method and computer pathological image analysis system.(3) Mitochondrial membrane potential (MMP) of cerebral neuron and percentage of cerebral neuron apoptosis were assayed by flow cytometry (FCM).(4) Change of neuron density of cerebral cortex and hippocampus was assayed by computer pathological image analysis system.(5) Serum interleukin-2(IL-2) was assayed by radioimmunossay(RIA).(6) Morphological change of cerebral cortex and hippocampus tissue was observed by photomicroscope.Result1. Compared with the normal control group, capability of learning and memory of model control group decreased substantially. Structural changes of cerebral cortex and hippocampus coincide with aged mice, which suggests that mice injected by D-gal suffer a manmade aging change of brain.2.
