STUDIES ON THE ACTIVE CONSTITUENTS OF Trachelospermum jasminoids AND Ervatamia Hainanensis Life sciences are focus on drug abuse, malignancy and virosis now. It is efficientand economic to obtain therapeutic compounds or leading compounds from traditionalChinese medicine. In this study chemical constituents isolation and identification ofTrachelospermum jasminoides (Lindl.) Lem and Ervatamia hainanensis Tsiangwere systematically performed. The anti-drug activity of total alkaloids of E.hainanensis was tested. The anti-tumor and anti-HIV activity test of some compoundswere also performed. The stems and leaves of T. jasminoides (Apocynaceae), known as "Luoshiteng"in Chinese folk medicines, have a history of use in the treatment of rheumaticarthralgia, aching of the loins and knees, and traumatic injuries. A number offlavonoids, lignans, and alkaloids had been discovered from this plant before.Furthermore, trachelogenin and actigenin, the main components of this plant, werereported to show strongly inhibit strain HIV-III B in vitro. In order to isolate morelignans, we systematically investigated the chemical constituents of it. By many kinds of separation methods, 34 compounds were isolated from freshstems and leaves of T. jasminoids. Their structures were elucidated based on spectradata (1H-NMR, C-NMR, DEPT, H-1HCOSY, NOESY, HMQC, HMBC, EI-MS, 13 1ESI-MS, IR). They were: trachelogenin(TJ01); arctigenin(TJ02); tracheloside(TJ03); trachelogenin4′-O-β-gentiobioside(TJ04); nortracheloside(TJ05); nortrachelogenin 8′-O-β-D-glucoside(TJ06*); matairesinol 4,4′-biglucoside(TJ07); trachelogenin amide(TJ08*);nortrachelogenin 5′-C-β-glucoside(TJ09*); tanegoside B(TJ10); trachelosperosideB-1(TJ11); trachelosperoside D-1 (TJ12); trachelosperoside F (TJ13*);trachelosperoside E-1 (TJ14); 3-O-β-D-glucopyranoside quinovic acid(TJ15);3-O-β-D-glucopyranoside quinovic acid 27-O-β-D-glucopyranosyl ester(TJ16); 3第二军医大学博士学位论文 ABSTRACT3-O-β-D-glucopyranoside cincholic acid 27-O-β-D-glucopyranosyl ester(TJ17);trachelosperogenin B(TJ18); apigenin(TJ19); apigenin 7-O-β-glucoside(TJ20);apigenin 7-O-β-neospher -oside(TJ21); luteoloside(TJ22); naringin(TJ23); 6,8-di-C-β-D-glucopyanosyl -apigenin(TJ24); roseoside(TJ25); icariside B5(TJ26);tracheloionoside(TJ27*); actinidioionoside(TJ28); sodium ferulate(TJ29); salicylicacid (TJ30); trachelinoside (TJ31*); vanillic acid(TJ32); benzenyl methanolglucoside(TJ33) ; dambonitol (TJ34). Among them, compounds TJ06,TJ08,TJ09,TJ13,TJ27,TJ31 are newcompounds, and TJ11-18,TJ23-26,TJ28-30,TJ32-33 were first isolated from thisplant. E. hainanensis, called Hainan Gou Ya Hua and Du Gen Mu in Chinese, is ashrub distributed in the southern part of China. The roots and leaves of the plant havebeen used in Chinese folk medicine for the treatment of stomachache, dysentery,snake-bites, rheumatic arthritis, hypertension and virus hepatitis. Many ibogaine-typeindole alkaloids had been isolated from this plant. Total alkaloids of it showedsignificant anti –drug effects. To investigate the effective components, we studied thechemical constituents of the root cortex of it, and 14 compounds had been isolated andidentified. They were: vobasine(TE01); coronaridine(TE02); 3-(2-oxopropyl) coronaridine (TE03);3-hydroxy coronaridine(TE04); 3-(2-oxopropyl) ibogamine(TE05)*; 3-(2-oxopropyl)ibogamine hydroxyindolenine(TE06)*; tabernamine(TE07); cycloartenol(TE08);β-amyrin acetate(TE09); (+) isolariciresinol 9-O-β-D-glucopyranoside(TE10);octacosanoic acid (TE11); hexatriacontanoic acid(TE12); β-sitosterol(TE13);daucosterol(TE14) Among them, compounds TE 05,06 are new compounds, and TE03,04,08-14were first isolated from E. Hainanensis.. The anti-drug activity of to...
