| Objective: To observe the clinic curative effect of Zishenhuaxian Yin treating fore-hepatocirrhosis, study the effect of Zishenhuaxian Yin on the experimental hepatic fibrosis of rats, and to discuss its mechanism that resisting hepatic fibrosis, we may provide new theoretical guidance for treating fore-hepatocirrhosis by using Chinese traditional medicine.Methods: 1) Taking 50 patients who suffer from clinic fore-hepatocirrhosis as objects, and taking orally Zishenhuaxian Yin prescribe for 6 months. Before treatment, recording clinical symptoms as fractions standard, and testing their indexes of curative effect, for example, the indexes of hepatic function, including ALT, AST, TBIL, ALB, A/G,PT%; the serum four items of hepatic fibrosis (HA, LN, PCIII, IV-C); the physical test of liver, gall bladder and spleen and the security indexes include the regulations of blood, urine and stool, electrocardiogram, kidney function etc. The above indexes would be tested again after 6 months, and compared with 30 patients who taking orally Dahuangzhechong capsule. 2) The wistar rats in which hepatic fibrosis model had been induced by carbon tetrachloride (CCL4) were divided into three groups at random. Model group: the rats were fed forage for eight weeks after CCL4 had been given for eight weeks; Preventive group: they were taked Zishenhuaxian Yin for eight weeks when being made model and then were fed forage for eight weeks; Therapeutic group: after the models weremade successfully, the rats were given Zishenhuaxian Yin for eight weeks. All rats were put into death at the 16~th . After treatment, testing the serum indexes of hepatic function including ALT, AST, ALB, GLB, A/G; testing the activity of SOD and the content of MDAin serum; testing the hepatic fibrosis indexes (HA, LN, PCII1,IV-C). Observing hepatic histopathology: denaturation degrees of hepatic cells were calculated by histological examination; Van Gieson was also performed, by which can calculate both the semi-quantity and variation of accumulation and dissociation of collagen. Observing the synthesis and expressions of Desmin, TGF—pi, Collage I , Collage III and MMP-1/ TIMP-1 in the liver tissue by immunohistochemical assay.Results: 1)6 months later, the scores declined, except the liver palm and Hypochondriac mass (P>0.05), the rest symptom's changes were better than before(P <0.05). Every indexes of hepatic function were improved obviously, the serum contents of ALT, AST, TBil were descend obviously(P O.01), and the rate of ALB and PT% was higher (P <0.05). the serum contents of hepatic fibrosis indexes were obviously lower than before (P <0.01), the change of HA was the most obvious, LN and PCIII second to that, and IV-C was the lowest. The show of B-ultrasonic was better after therapy, the integrals of total, liver, spleen and portal vein were lower than before. There were significant difference in those items compared with before it was treated (P <0.01). After statistics, the improvement of every indexes in therapeutic group was better than contral group of Dahuangzhechong capsul,and there was significant difference (P<0.05, P<0.01) <> 2) After cured by Zishenhuaxian Yin, the indexes of hepatic function have improved obviously, and improvement of preventive group was better than therapeutic group; To the lipid-peroxidation, therapeutic group and preventive group can increase the activity of SOD and decrease the content of MDA in serum, which there were significant difference with model group(P<0.01). Compared with therapeutic group, the change of preventive group was a bit better; The contents of HA, LN, PCIII, IV-C were lower in given medicine groups than those in model group(P<0.05, PO.01). We may find that the groups given with Zishenhuaxian Yin can reduce markedly the degrees of hepatic cell denaturation through light lens. By compared with model group, the integral calculus has significant difference (P<0.05, P<0.01). Their integral calculus with semi-quantity were lower than model group (PO.05, P<0.01), but there was no difference between therapeutical and preventive group. The expressions of Desmin , TGF-P 1,Collage I ,CollageIIl and TIMP-1 measured by immunohistochemical assay in medicine groups were less than model group , the expressions of MMP-1 and MMP-1/TIMP-1 ratio were more than model group (PO.05, PO.01).
