Active portion were obtained from Er-xian decoction (EXD), bio-assayed with phannacological activities on the osteoblast and the osteoclast in vitro model. 22 compounds were obtained from the active portion. In order to investigate the pharmacological effect and principal mechanism of antiosteoporosis in EXD, several screening models including OVX as well as cell cultivation in vitro were used to evaluate their effectiveness. Our results clearly indicated the mechanism of action in EXD and active mass for antiosteoporosis. The present data confirmed that most compounds of the active mass had significant effects on antiosteoporosis by promoting proliferation of osteoblast and inhibiting bone resorption. The quantitative determination of the total flavonoids, alkaloids and saponins in EXD were carried out by the way of UV spectrophotometry. The RP-HPLC method was established for quantitative analyses of active components in EXD. Meanwhile, the quantitative analyses of the active components including its typical compound in EXD were determined by RP-HPLC method. Dynamic content changes of active compounds in crude drug and formula were studied. The best extract technology and the purification conditions by absorption resin were optimized.
