BackgroundThere are a variety of symptoms occur during women menopause in addition tomenstrual disorders, say, waves of hot sweating, irritability, upset, insomnia,urinary incontinence, And for the most part cardiovascular disease(Cardiovascular disease, CD) is the leading cause of diseases and death inpostmenopausal women, which accounts for about 30%. It has been revealed thatEstrogen (estrogen, EST) is closed to the occurrence of female CD, but effects ofEST on CD prevention and treatment for women after menopause is inconclusive.Clinically, Chinese medicine has a better CD control role for postmenopausalwomen, while its mechanism is not clear. Therefore, the research upon themechanism of Chinese herb to improve the health of women is of greatsignificance.In ancient Chinese medicine works, there is no disease term corresponding directly to menopause syndrome. Not until the 20th century, Chinese gynecologistZhuo Yunong referred to "all evidence before and after menopause". In theChinese classics, many discussed pathogenesis, differentiation, and medicinalprescriptions regarding menopause syndrome. Therefore, to sum up and toanalyze the ancient books not only help to understand the theory of femalemenopause syndrome, but also help to further improve the efficacy of traditionalChinese medicine.In this study, based upon 32 kinds of ancient works, we summed up the relevantdiscussion of the menopausal syndrome and observed the functions of Erxiansoup upon ovariectomized rats, so as to explore initial mechanism of action of thecompound.ParPart 1 The summary of the menopausal syndrome inChinese Ancient BooksObjective:To summarize ancient works with respect to pathogenesis,differentiation, and Medicinal Prescriptions of menopausal syndrome.Methods:Reading and summarizing the 32 original texts, including "YellowEmperor", "hard by the Yellow Emperor eighty-one" "Febrile Diseases,""Synopsis of Golden Chamber" and the like.Results:The "Yellow Emperor" described the most comprehensive in the theoryof menopausal syndrome, involving the kidney, menstruation, Chong Ren,menstruation, etc.; Later ancient physicians discussed most about the menstrualdisorders, and the mechanism varied from blood heat, blood cold , Liver and spleen deficiency to liver qi stagnation. The prescriptions involved Si-Wu-Tang,Ba-zhen-soup, Xiaoyaosan and warm soup,etc.ConclusionConclusion: The theory of "Yellow Emperor" with regard to the menopausalsyndrome is the backbone for future treatment. The treatment has gone throughthree different stages, from the blood of governance, to the liver and spleen ofgovernance, ending up to the blood , Liver, spleen and kidney of governance. Theprescriptions mainly included Si Wu Tang, Xiaoyaosan and Dang guisan.Part 2 The effects of modified ER-Xian Decoction on E2and the vasomotor of ovariectomized ratsObjective: To observe the effects of modified ER-Xian Decoction on sexhormones, vascular contraction and relaxation, vascular structure and blood lipidsin ovariectomized rats.Methods: 56 adult female SD rats were randomly divided into 7 groups: controlgroup, sham operation group, model group, OVX + Decoction of high, mediumand low dose group (Chinese high, medium and low-dose group), OVX + tibolonegroup (tibolone group). the change of body weight, rat tail artery blood pressure,abdominal aortic relaxation and contraction, structural changes in the abdominalaorta, immune serum estradiol (E2), luteinizing hormone (LH), follicle stimulatinghormone (FSH) levels and lipid changes were observed before and after theinterventions. ResultsResults: Compared with model group, Chinese medicine can significantlyincreased serum E2 levels (P <0.05), and significantly low FSH, LH, body weight,systolic blood pressure (SBP), diastolic blood pressure (DBP) levels, totalcholesterol (TC ) And triglyceride (TG) levels (P <0.05), in which high dosegroup was the most significant; medicine high dose group rats significantlyimproved the abdominal aorta, blood vessels systolic, diastolic function andvascular structure, which was no significant difference with tibolone. Each groupHDL and LDL were no significant difference (P >0.05). Conclusion: Themodified ER-Xian Decoction can reduce body weight, blood pressure and bloodlipids in ovariectomized rats, and meanwhile, improve vasomotor function andvascular structure. Its mechanism may be related to the increased E2 level.
