Insulin resistance refers sensitiveness and reactivity of target tissue to Ins degrade, and is indicated for all kinds of clinic manifestations which are caused by reactivity decreasing of target tissue cell such as liver, muscles and fat to insulin biological effect, combined with hyperinsulinemia leading to all kinds of diseases such as diabetes, lipid metabolism disorder, hypertension. The investigation aims at elucidating active site and mechanism of action that American ginseng treats insulin resistance, so provides the experiment foundation for American ginseng's application in insulin resistance relative diseases such as diabetes, hypertension and so on.Objective: sieve rat insulin resistance' s active site that American ginseng treats high fat diet, debate active site's mechanism of action to treat insulin resistance.Methods: 1 Apply macroporous resin to enrich American ginseng's different pharmaco-using location2 Regard wistar rat as the experimental object, apply the animal model of rat insulin resistance to sieve active site on the different pharmaco-using locations, and divide rat into seven groups, which are mould group, normal control group, medicine group, water elution fraction group, 30 percent alcohol elution fraction group, 60 percent alcohol elution fraction group, 95 percent alcohol elution fraction group, determine blood sugar, insulin and calculate insulin's sensitive index, regarding insulin's sensitive index as evaluation index.3 Observe the active site's influence on pancreatic gland morph, apply immunohistochemical method to observe active site's influence on insulin receptor, employ PT-PCR method to observeactive site's influence on insulin receptor mRNA.4 Observe the active site's influence on cholesterin and triglyeride. Employ radioactivity immunity method to observe the active site influence on fat cytokine LEP, TNF- a . Conclusions: 1 The active sites of American ginseng treating insulin resistance are 30 percent alcohol elution fraction and 60 percent alcohol elution fraction.2 American ginseng's active site can obviously improve insulin resistance rat's state, degrade insulin on an empty stomach, and improve insulin sensitivity.3 American ginseng's active site can obviously improve pancreatic pathologic state, strengthen liver cell envelope insulin receptor's express, so improve insulin resistance from insulin receptor link.4 American ginseng's active site implements improving insulin resistance from insulin receptor link through strengthening insulin receptor mRNA's express.5 American ginseng's active site can improve TNF- a and Lep's express, improve insulin resistance from the view of fat metabolism and fat cell factor.
