Asthenia cold syndrome belong to the theory of TCM syndrome of cold syndrome,also known as Yang deficiency syndrome.The pathogenesis of Asthenia cold syndrome,namely the sun be the spirit loss,body temperature raising,the action of gasification is abate,too much Yin qi,declining qi,blood,viscera function in body.Modern medicine theory think that Asthenia cold syndrome body functions is poor then cause asthenia body,mainly function abate,such as lower metabolism function,adrenal function is low,etc.TCM holds that American ginseng has tranquilizing and allaying excitement,boosting Qi and nourishing Yin,clearing heat and promoting fluid effect and so on,the modern pharmacological studies have shown that it has antithrombotic,anti-arrhythmic,shock resistance,stress resistance,anticancer,reducing blood fat,strengthen the body immunity and protect liver function and so on.American ginseng slightly bitter,taste sweet,cold,belongs to cold medicine.In cold individuals or in winter will appear abdominal pain,sometimes cold sometimes hot,limbs cold symptoms,such as limits the widely application of American ginseng.This study to join the warm biased raw ginger for American ginseng processing,get Processing American ginseng,improved the American ginseng slants cold sex,at the same time enhances the American ginseng yiqi,taking the effect of vital qi and dryness.Experimental objective:This study establish Asthenia cold syndrome in rats model,to explore the effect of American ginseng and Processing American ginseng in Asthenia cold syndrome rats and its mechanism,the influence of provide experimental basis for its exploitation and utilization.Experimental method:112 Wistar rats were randomly divided into eight groups,namely: the normal group,model group,American ginseng of small,medium and large dose groups,Processing American ginseng of small,medium and high dose group.Each group has 14 rats.except the normal group,other groups intragastric administration cold medicine 4 ml,one times a day,continuous dosing 35 days,normal group intragastric administration amount of distilled water.on the day of 8,the normal group and model group give 0.5% sodium carboxymethyl cellulose,American ginseng of small,medium and large dose group give American ginseng respectively 0.5,1.0,2.0g/kg,Processing American ginseng of small,medium and large dose group give respectively Processing American ginseng 0.5,1.0,2.0g/kg.one time per day,for 28 consecutive days,delivery volume for 10ml/kg.monitor rats body weight,heart rate every week.After 28 days,detect the rat gastric emptying rate,urine 17–OHCS,E,NE and DA content,Serum TSH,c AMP,cGMP,IL-2 and IL-8 content,calculate the c AMP/cGMP ratio,Stomach tissue c AMP,cGMP content,calculate the c AMP/cGMP ratio,Optical microscope stomach tissue morphology change.Experimental result:1.Rats in normal group eating and defecate is normal,fur luster,normal behavior,Model group,American ginseng,1.0,2.0g/kg rats appeared runny stool,diarrhea,filthy fur near the hips,lazy to move,fur is loose and matt,Processing American ginseng in each dose group and defecate normal diet,hair luster,behavior is normal,same as the normal group.2.Compared with normal group,model group and American ginseng every dose group of rats 1~4 weeks there is a downturn in the weight Processing American ginseng every dose group of rats 1~4 weeks no difference between the weight.Compared with model group,American ginseng every dose group of 1~4 weeks of rat were no obvious difference of weight,Processing American ginseng 0.5,1.0g/kg can make the 3,4 weeks of rats body weight increased significantly(P<0.05 or P<0.01),2.0g/kg can make 2~4 weeks of rats body weight increased significantly(P<0.05 and P<0.001).3.Compared with normal group,model group and American ginseng every dose group of rats 1~4 weeks heart rate slowed significantly(P<0.05 and P<0.001),Processing American ginseng 0.5g/kg 1~3 weeks rats heart rate slowed significantly(P<0.05 or P<0.01),1.0g/kg 1,2 weeks rats heart rate slowed significantly(P<0.05),2.0g/kg week 1 heart rate slowed significantly in rats(P<0.05),no difference between the 2~4 weeks.Compared with model group,American ginseng dose groups each rat heart rates did not differ significantly 1~4 weeks,Processing American ginseng each dose group of 2~4 weeks rats heart rate has a rising trend.4.Compared with normal group,model group rats urine 17-OHCS,E and NE and DA content were significantly lower(P<0.05 and P<0.001),American ginseng every dose group of rats in the urine 17-OHCS,E and NE content were significantly lower(P<0.05 and P<0.001),Processing American ginseng 0.5g/kg NE content in rats significantly decreased(P<0.05),Compared with model group,American ginseng,2.0g/kg in rats 17-OHCS,E and NE content were significantly lower(P<0.05 or P<0.01),Processing American ginseng 1.0g/kg 17-OHCS and NE content in rats were significantly increased(P<0.05),2.0g/kg in rats 17-OHCS,E and NE content were significantly increased(P<0.05 or P<0.01).5.Compared with normal group,model group and American ginseng each dose group of rats serum content of c AMP and the c AMP/cGMP ratio were significantly lower(P<0.05 or P<0.01),Processing American ginseng each dose group of rats serum content of c AMP and cGMP and c AMP/cGMP ratio has no obvious difference,Compared with model group,American ginseng every dose group of rats serum content of c AMP,cGMP and c AMP/cGMP ratio has no obvious difference,Processing American ginseng 1.0,2.0g/kg of rat serum content of c AMP and the c AMP/cGMP ratio were significantly increased(P<0.05 or P<0.01).6.Compared with normal group,model group rats TSH and IL-2 levels were significantly lower(P<0.05),no difference between the content of IL-8,American ginseng every dose group of rats TSH levels significantly decreased(P<0.05 or P<0.001),IL-2 and IL-8 content has no obvious difference,Processing American ginseng each dose group of rats serum TSH,IL-2 and IL-8 content had no obvious difference,Compared with model group,American ginseng,2.0g/kg of rat TSH decreased obviously(P<0.05),American ginseng Jiang Mi system 1.0,2.0g/kg of rat TSH and IL-2 were significantly increased(P<0.05 or P <0.01).7.Compared with normal group,model group rats stomach tissue c AMP,the content and the c AMP/cGMP ratio significantly decreased(P<0.01),American ginseng and Processing American ginseng each dose group of rat stomach tissue c AMP and cGMP content and c AMP/cGMP ratio has no obvious difference,Compared with model group,American ginseng dose groups each rat stomach tissue c AMP and cGMP content and c AMP/cGMP ratio has no obvious difference,Processing American ginseng 0.5g/kg of rat stomach tissue c AMP content significantly increased(P<0.05),Processing American ginseng 1.0,2.0g/kg of rat stomach tissue c AMP content and the c AMP/cGMP ratio were significantly increased(P<0.05 or P<0.01).8.Compared with normal group,model group rats gastric emptying rate significantly decreased(P<0.05),American ginseng and Processing American ginseng each dose group no obvious effect on gastric emptying rate of rat,Compared with model group,American ginseng every dose group of Processing American ginseng 0.5,1.0g/kg for rat gastric emptying rate has no obvious influence,Processing American ginseng 2.0g/kg of rat gastric emptying rate increased significantly(P<0.05).9.Histopathological detection: normal group rats stomach four layers structure is clearly visible,mucous membrane layer is the most thick,gastric mucosa is arranged closely,mucosal epithelial cells are tidy and uniform,at the bottom of the stomach main cells and wall cells is clearly visible,gastric submucosa,muscular layer and adventitia was inflammatory cell infiltration and vascular engorgement of hemorrhagic change.Model group,American ginseng and Processing American ginseng each dose group compared with normal group had no significant difference.Experimental conclusion:1.American ginseng can increase behavior Asthenia cold syndrome rats appeared abnormal state,can make the body weight,heart rate slows,Processing American ginseng can make the Asthenia cold syndrome rats behavior status,body weight and heart rate gradually returned to normal.Prompt American ginseng aggravate the Asthenia cold syndrome in rats.Processing American ginseng have improved significantly.2.American ginseng can significantly reduce the Asthenia cold syndrome in rats urine 17-OHCS,E and NE content and Processing American ginseng can significantly increase the Asthenia cold syndrome rats urine 17-OHCS,E and NE content.American ginseng can reduce rats by sympathetic-adrenal medulla function,increase the cold symptoms,Processing American ginseng can strengthen Asthenia cold syndrome rats sympathetic-adrenal medulla function,it may be to improve one of the mechanisms of cold symptoms.3.American ginseng can reduce Asthenia cold syndrome rats serum TSH levels,Processing American ginseng can significantly increase the Asthenia cold syndrome rats serum and stomach tissue c AMP content and c AMP/cGMP ratio,increase the serum TSH levels.American ginseng can make the body substance and the decline in energy metabolism,increase the cold symptoms,Processing American ginseng by improve the ability of the body's metabolism,increase energy metabolism Asthenia cold syndrome rats,so as to improve the cold symptoms.4.American ginseng of Asthenia cold syndrome rats serum IL-2 content had no obvious effect,Processing American ginseng can significantly increase the Asthenia cold syndrome IL-2 content in serum of rats.Asthenia cold syndrome antagonism the action of American ginseng of immune enhancement and made Processing American ginseng effect still exists,it improved the rats of cold symptoms.5.Under this model between groups were not seen stomach tissue pathology morphology change,explain the rat experimental Asthenia cold syndrome and participation in the Processing American ginseng and American ginseng has not been morphology damage.
