In this study, with the research model of"components of TCMs", centering on constructing multi-components library (MCL), methods of chemistry and biochemistry were carried out to elucidate the substantial foundation of TCMs. Kudzu root was selected as the target of investigation and methods were discussed for separation and characterization.For the bioactive characterization of MCL, recombinant yeast screening assay (YES) was developed for the screening of phytoestrogens. The estrogenic activities of the crude extract and fractions with different polarities of kudzu root were preliminarily investigated.A stable, reliable and reproducible HPLC method was developed for the analysis of kudzu root, a complex system. The HPLC model, mobile phase and additive were optimized. The HPLC retention parameters a, c and peak shape parameters sigma, tau were obtained correctly and quickly by using five linear gradients. Then CSA software was used to simulate the gradient conditions in experiments and the optimized condition were obtained. The crude extract and fractions with different polarities of kudzu root were characterized by HPLC-UV-DAD under the optimized gradient condition. The data from HPLC analysis and bioactive screening were correlated. The crude extract of kudzu root was analyzed by HPLC-APCI-MS.29 and 18 components separated from ethyl acetate and n-butyl alcohol fractions respectively were obtained by preparative HPLC and relative techniques.
