Study of controlled release drug is one of the hot spots in the field of pharmaceutical research, its purpose is to look for a way to provide an ideal drug concentration in blood, and improve the safety and availability of drug. The controlled release membrane is a dosage form prepared through the method of coating drug pellets using polymer materials, and has properties such as stable release velocity, ripe preparation crafts. The polymer material controlled release membrane and drug pellets are the key of controlled release effects, their composition and property will affect the release velocity of controlled release membrane. At present, the study of preparation, properties, and influence factors of polymer material controlled release membrane was very active.The study's expected objectives is to prepare controlled release membranes of anti-tumor drug Lomustine(CCNU), Vincristine(VCR) using collagen protein as polymer material carrier, then planted into the tumor of cancer patients after operation, to achieve the goal of constant drug release.We tried different preparation crafts to prepared CCNU microcapsules, VCR microspheres by recoacervation and drying-from-oil methods individually, then mixed them into collagen swelling solution to become membrane by freeze drying. The distributions of diameter are 0-3.5μm 20.9%, 3.6-39μm 74.8, >40μm 13.6% (CCNU), 0-50μm 20.9%, 51-298μm 76.3%, >298μm 2.8%(VCR); Ratio of encapsulation was 65.7%(CCNU), 59.1%(VCR) determined by ultraviolet spectrophotometry; Drug content in tumor tissue of conventional administration and release velocity of membrane invitro were determined by HPLC. After above-mentioned determination, we defined the contents of drugs put in the micropellets were 12. 2mg(CCNU), 1.11mg(VCR).
