Background:We have previously done the study"screen the genes related to mouse brain development and injury by high density cDNA microarrys", In which we found 560 genes high expressed, 34 genes overexpressed in mouse development. These genes could be divided into 17 classes, functions varied from regulation of transcript, signal transduction, cell metabolism, immune regulation, cytoskeleton, DNA replication repairing, cell adhesion and so on. Remarkably, gene BC023882 expressed high level in the brain only embryo and postnatal nonage, which function is unclear. Relative gene expression ratio (Cy5/Cy3) was 7.6038 on P0 day, much higher than the standard of screening differentially expressed gene(ratio>2.0000), which suggested gene BC023882 may play an important role in central nerve system(CNS) of mouse development.Methods and Results:1. Express pattern and brain localization of gene BC203882(1). Semi-quantitative PCR to detect express pattern of gene BC023882 Semi-quantitative PCR following reverse transcription was used to assess transcript levels of the gene BC023882 in various tissues, including embryo (brain, body), brain, heart, lung, kidney, liver and intestine of E14 d,E19 d,P1 d,P7 d,P14 d,P30 d and P60 d Kunming mouse. The mRNA of gene BC023882 transitorily expressed in mouse development. The highest expression level of gene BC023882 was present in brain on few antenatal and postnatal days. The gene BC023882 were weakly expressed in mice both embryonic body tissue and liver of early postnatal days, other tissues of experiment groups were not detectable.(2) In situ hybridization to detect mRNA of gene BC203882 in mouse brain Digoxygenin labeling cRNA probe preparation by in vitro transcription PcDNA3.1(-) constructed with the gene BC023882 cDNA insert was linearized and transcribed in vitro from the T7/T3 polymerase promoter to yield the cRNA probe.
