[OBJECTIVES]Myelodysplastic syndromes (MDS) are a heterogeneous group of malignant clonal stem cell disorders characterized by peripheral blood cytopenia, dyshematopoiesis with hypercellular bone marrow. Patients with MDS had poor responses to most therapeutic modalities and bad prognosis owing to a high risk of progression to acute myeloid leukemia (AML). As a preleukemic disorder, MDS was a good model to be studied the step-by-step mechanisms of preieukemia transformation into overt leukemia. In the process of transformation, little was known about the cell cycle regulation in association with malignant transformation. So we analyzed cell cycle and cell cycle control genes expression in MDS, AML evolving from MDS (MDS-AML), primary AML and normal control.[METHODS]1. The analysis of cell cycle was adopted by propidium iodide stain to examine the cell cycle phases (GO/G1, S and G2/M) on bone marrow mononuclear cells (BMMNCs) from 22 normal, 27 patients with MDS, 4 MDS-AML and 12 primary AML.2. Immunofluorescent double stain was conducted to show the rate of Ki67 positive cells (proliferate cells including G1, S, G2 and M phase) on BMMNCs and on CD34~+ cells from 16 normal, 26 MDS, 4 MDS-AML and 16 primary AML.3. BMMNCs from eight normal and 15 MDS were plated at 5 × 10~5 cells/ml in IMDM supplemented with 20% FCS and 100U/ml penicillin/streptomycin and were cultured at 37℃, 5.0%CO2 and 95% humidity in 6-well culture plates with different growth factors cocktail.
