Obesity has become a global health problem, and it is linked to the development of many angiogenesis-related diseases such as diabetes, cardiovascular disorder and cancer. Adipose tissue serves as an energy storeage depot as well as secretary tissue which secretes a wide range of adipocyte-derived biologically active polypeptides termed adipocytokines. Adiponectin (ACRP30, ADIPOQ, apMl gene product) is a recently discovered protein secreted exclusively by adipocytes that acts as an antidiabetic and antitherogenic hormone. Circulating levels of adiponectin are reduced in conditions related to insulin resistance and hyperinsulinemia, such as visceral obesity and type 2 diabete.Very recently, some studies found evidence that adiponectin was inversely related to the risk cancers. Additionally, recent studies in vitro have demonstrated that adiponectin inhibits endothelial cell proliferation and migration, in the chick chorioallantoic membrane and the mouse corneal angiogenesis assays, adipoenectin remarkably prevents new blood vessel growth, and adiponectin serves as a negative regulator of angiogenesis. In another study, adiponectin has been reported to stimulate the differentiation of human umbilical vein endothelium cells into capillary-like structures in vitro and stimulate blood vessel growth in vivo, the study indicates that adiponectin serves as a positive regulator of angiogenesis. As limited evidence is available concerning the possible role of adiponectin and the potential impact on angiogenesis and tumor. To provide additional information on the issue, we have conducted this work, our study mainly focuses on two aspects below: 1. Cloning and expression of ACRP30 and globular ACRP30 to get purified recombinant ACRP30. 2. The influence of recombinant ACRP30 on cell proliferation and angiogenesis.The methods adopted in this study:Total RNA was isolated from mouse adipose tissue using Trizol reagent according to the manufacture's instructions.
