| Objective :To observe the microvascular pathological change of diabetic rats and the protective effect of JiangTangTongMaiFang on the diabetic microvascular complications in the diabetic rats, and investigate the molecular pathogenesis of the protective effect of JiangTangTongMaiFang on the diabetic microvascular complications.Methods:1 Wistar rats were induced for diabetes by streptozotocin intravenous injection and treated with high-fat-glucose chow.2 Experimental rats were devided into 4 groups: Chinese medicinal materials group(group of JiangTangTongMaiFang)/western medicine group(metformin group)/ model group/ normal control group.3 After 8 weeks'treatment, the percentage of CD54 positive cells in fresh whole blood was detected by flow cytometry, the function of pancreatic island was evaluated by sugar tolerance test, and the plasma levels of SOD, MDA, HDL, TG, FFA, NO, ET, vWF, Cr and the urinary ALB concentration were measured.4 The tissue of the living rats was kept for detecting the expression of VEGF. type 1 collagen and laminin in kidney by immunohistochemistry (IHC) , detecting the expression of PAI-1mRNA/TGF- β 1mRNA by in situ hybridization histochemistry (ISHH), and observing the kidney ultramicrostructure change under light and electron microscope .Results:1 Test one showed that:(1) Body weight(BW) and blood glucose of model group increased ,and it indicated... |
