The Study Of Protective Effect Of Tang Mai Ning On Diabetic Angiopathies In Diabetic Rats

Objective :The present study has been undertaken to declaim the macrovascular pathological change of diabetic rats, and the protective effect of TangMaiNing(TMN) on the diabetic angiopathies in the diabetic rats, and investigate the protective pathogenesis .Methods:1 Aminal model:Sixty adult wistar rats (SPF, 200 g-220 g body weight, female) , abtained from the deasease control center of huibei province, these animals were normally fed with water and chow at room temprerature in a light/dark cycle of 12/12 hours. After 1 week, they were devided into two groups: the mormal control grouP (n=10) and the modol group(n=50), the modol group were induced in diabetic rats with 0.9%alloxan by injecting into the caudal vein at 45 mg/kg. At the 2th and 5th hour, 25%glucose intragastric at 2 ml/100g. Then,measured the random blood sugar(RBS) concertration at the 3th,7th,14th,21th day, while the RBS≥ 10 mmol/L, the rats were concerded as diabetic rats. In the end, the number of diabetic rats were 38, the normal control were 16.2 Groups and treatment: the rats were devided into 5 groups : TMN(shengdihuang maidong wuweizi zhimu tianhuafen xuanshen danshen)group, YouJiangTang(YJT) group, (TMN+YJT) group, model group, normal group. TMN group were treated with 1.25 g/ml TMN 2ml, YJT group were treated with 3.87 mg/ml YJT 2ml, TMN+YJT group were treated with 1.25 g/ml TMN 1ml and 3.87 mg/ml YJT 1ml, model group were treated with 0.9% Nacl 2 ml, normal grouP were treated with 0.9% Nacl 2 ml, these rats were intragrastriced once a day, total 4 weeks.3 Biochemical measurement: we investigated the clinical changes, to fast over 8 hours, 0.5% pentobarbital at 30 mg/kg were injected into the abdominal cavity, get blood from heart, the blood samples were separately saved in centrifuge tube with heparin anticoagulation or EDTA-Na2 solution, centrifuged at 3000 rPm/min 10 minitues, collected the Plasma, saved in-20℃. We measured the glucose(GLU), triglyceride (TG),total cholesterol(TC), insulin(INS), P-selectin, nitric oxide(NO), endothelin(ET) concentration in Plasma.4 HE staining: biopsied the rats, the tissue of macrovascular were kept for hematoxylin-eosin(HE).
