Caulophyllum robustum Maxim which belongs to the family of Berberidaceae is widely distributed in Shanxi province,Gansu province,Sichuan province,the northeast of china and the far east of Russia. The officinal part of the plant was roots and rhizome which can cultivated. According to "Chinese traditional medicine dictionary", the herb is a folk medicine that has been utilized widely to cure many diseases such as rheumatoid arthritis, stomachache and bellyache,fracture injuries and menstruate disorder etc. due to its strong and wide biological activities. The chemical constituents of the plant were alkaloids and triterpenoid saponins according to literature.Firstly,The results of pharmacological experiment results confirmed that the activepart of anti-rheumatism. Then, chemical and pharmaceutical research were conductedon the effective part for the aims of further determining the active compounds and the effect mechanism for anti-rheumatism. 35 compounds were isolated with sil-gel, r eversed-phase (ODS) column chromatography and PLC, of which the structures of 32compounds were fully determined according to the chemical analysis and spectrosco pic evidences (1D and 2D-NMR). Among them there are 24 pentacyclic triterpenoid saponins compounds, 2 alkaloids compounds, 1 sesquiterpenoids compound, 3 sterols compounds, 1 iridoids compound and 1 aromatic compoud. 9 novel compounds werereported and 14 compounds were firstly found in the Caulophyllum genus and 16 compouds were firstly found in the Caulophyllum robustum M. The structures of thenine novel compounds were identified as leiyemudanoside A[3-O-α-L-arabinopyranos yl-hederagenin- 28-O-β-D -glucopyranosyl -(1→6) -P-D-glucopyranosyl ester], leiyemu danoside B [-O-β-D-glucopyranosyl - (1→3) -α-L-arabinopyranosyl -caulophyllogenin-28-O-α-L-rhamnopyranosyl-(1→4) -β-D -glucopyranosyl -(1→6) -β-D -glucopyranosyl ester], leiyemudanoside C[3-O- β-D-glucopyranosyl- (1→2) -a-L-arabinopyranosyl -cau lophyllogenin-28-O-α-L-rhamnopyranosyl-(1→4) -β-D -glucopyranosyl -(1→6) -β-D -gl ucopyranosyl ester], leiyemudanoside D[3-O -α-L-arabinopyranosyl-caulophyllogenin- 2 8-O-α-L-rhamnopyranosyl-(1→4) -β-D -glucopyranosyl -(1→6) -β-D -glucopyranosyl-(1 →4) -a-L -rhamnopyranosyl -(1→4) -p-D -glucopyranosyl -(1→6) -β-D-glucopyranosy 1 ester], leiyemudanoside E[3-O-β-D-glucopyranosyl- (1→3) -a-L-arabinopyranosyl ~e chinocystic acid -28-O-α-L-rhamnopyranosyl -(1→4) -P-D -glucopyranosyl -(1→6) -β-D -glucopyranosyl ester], leiyemudanoside F[3-O-α-L-arabinopyranosyl -11-oxo-caulop hyllogenin-28-O-α-L-rhamnopyranosyl-(1→4) -β-D -glucopyranosyl -(1→6) -β-D -glucopyranosyl ester], leiyemudanoside G[3-O-a-L-arabinopyranosyl -ll-oxo-hederagenin-28-O-a-L-rhamnopyranosyl-(l--4) -P-D -glucopyranosyl -(1—6) -P-D -glucopyranosyl est er], leiyemudanin A [ (6S, 9R) 6-hydroxyl-3-oxo-a-ionol-9-O-p-D- glucopyranosyl-(l -~6)-a-L-rhamnopyra noside], leiyemudanin B [4— (p—O-P-D- glucopyranosyl) —ph enyl-3 -methyl crotonate ] oThe method of quality control was established by HPLC-ELSD with 5 triterpen-oid saponins as standards. It confirmed that the method was simeple ,accurate,effecti-ve and feasible.This thesis further confirmed the pharmacological action of effective part of Ca-ulophyllum robustum M. and the effective substances for the treatment of rheumatism. The results provided important scientific foundation for further reserch and the development and will promote the study on the effecting mechanism of Caulophyllum robustum M. against rheumatism.The combination of security, effectivity and controllability of the medicine was realized by the modern technological methods in the research.
