Research On Extraction And Isolation Of Red Component And Pharmacognostic Quality Evaluations From Caulophyllum Robustum

Objective:To optimize a technology to extract and purify the major component of the alkaloids and saponins from Caulophyllum robustum;And to prepare compounds of red components;To develope a HPLC method to control contents of saponins from Caulophyllum robustum,and to analysis the quality of the herb from different origins,years,seasonsand parts.Methods:Based on contents of six medicinal ingredients contents and dry extractives percent as an index,processing data by weighted average method;or the goral experiment was used to optimize the extraction method and four factors,ethano’s concentration,time,the amount of solvent and number of times,were chosen as the extraction process.We used the silica gel column chromatography method to separate marking red component of six compounds from CR under the different corditions.The experiments evaluated pharmacognostic quality based on the contents from such as different origin,seasons,years,parts.Results:Ten compounds were isolated from CR and identified as Caul oside C,Hederagenin,Oleanolic acid,Magnoflorine,N-methylcytisine,Cauophylline B,Cauloside H,Leonticine D,Cauloside G,Cauloside D.The cordition under A2B2C2D2 is 10 times 80% ethanol extracting for 2 hours.The experiment analyzed the contents from different origins,years,seasons and parts.Congsidering TAK,TSP contents,there were significant differences of Caulophyllum robustum from Heilongjaing,Hebei and Sichuan.In different months,there were significant differences of Caulophyllum robustum in June,July and September.There were significant differences of Caulophyllum robustum in 2012,2013 and 2014.TAK,TSP contents were significant differences in rhizome and fibrous roots.ConcLusion:We obtained representative component Caulophylline B and other nine compounds,In particular,compounds 2 and 3 were isolated for the first time.The best extraction process was found.Focusing on the TAK,TSP contents and TAK/TSP,Content with the increase of latitude increased after decreased first.September should be the best time,research on medicinal parts,the TAK are mainly in rhizome,while fibrous roots contain the most of TSP.
