Lung cancer is one of the maiigmancies with high incidence and mortality. But there is still lack of a noninvasive procedure for correct diagnosis and staging.PURPOSEThe purpose of this study is to improve lung radioimmunoimaging (RJI) including: the production of highly specific and sensitive anti-lung-cancer-cell monoclonal antibodies and their fragments, the selection of a suitable radionuclide for labelling, the application of new imaging techniques and the verification of our experimental studies by clinical trialsMETHODSExperimental study (1).2E3 ,an antibody against human large cell lungcancer formally raised in our laboratory was reprepared and its F(ab')2 wasalso produced. (2).Nude mice bearing human lung cancer PLA-801 (large cell cancer cell line) or Hu A-1(adenocarcinoma cell line) xenografts (3)I-125,1-131 and Tc-99m were used for labelling. (4) Three kinds of IgG monoclonal antibodies,2E3,Sm-1(anti lung aquamous cell cancer), Am-7(anti lung adenocarcinoma ceil) and one IgM monoclonal antibody (LC-l,anti lung adenocarcinoma cell,supplied by Shanghai Institute of Cell Biology) were labelled with radioiodine or technetium-99m and their immunoreactivities were studied by animal model imaging. The in vivo T/NT value was calculated dynamically using ROI technique and biodistribution was measured 24h after injection of the antibody.Clinical study From Oct,1993 to Nov,1995. RII was performed in 66 patients. Amomg them, 54 patients were studied with Tc-99m-labelled IgG monoclonal antibodies and 8 patients with IgM McAb fragments. In addition to imaging, SPECT technique was used in 33 patients also.
