The instence of congenital heart diseases and the present condition of cardiac surgery in our country make it necessary to find the procedures with which the development of the course of pulmonary hypertension (PH) can be reduced, and the reality of mountain-rich promote us to study the effects of hypoxia on the hyperkinetic pulmonary hypertension. The systemic study of the "plexogenic pulmonary arteriopathy" has some limitation, but can be approached by animal experiments.This study is to observe the effects of renin-angiotensin system on the base of establishment of the rat model of shunting, hypoxia and shunting plus hypoxia.At present, neither rat shunting PH model and shunting with hypoxia PH is reported in the world, nor the hunoral factors as renin-angiotensin system in shunting PH.1. The establishment of left-right shunting PH in ratsEighty healthy male Wistar rats, weighing 170-250 grams, were chosen as the subjects of experiment, fifty of which underwent operations by the use of sodium pentabarbita intrapericoneally injection.The chest was opened along the middle line of sternum, the left common carotid artery was anastomosed with the main pulmonary artery to form a systemic-pulmonary shunting. Survival rate of the rats after operation was 60% (30/50). Four, 8 and 12 weeks later after operation, the hemodynamics, morphologic changes of the pulmonary vasculature, were appreciated, and the plasma renin activity (RA), angiotensin I (AI) and II, and argine vasopressin (AVP) were measured. Nine of 24 (37.5%) shunting rats showed PH by the scheduled time, of which 5/18 (27.750 occured in the simple shunting groups. The pulmonary vasculature of the 3 rats in 12 weeks shunting group showed typical alterations,being I and II degrees of the Heath-Edwards grading system.2. Construction and effects of hypobaric hypoxiaThe hyperbaric sterilizer equipped with a pressure-autoregulator, in which the pressure was 400mmHg (vaccum pressure -360 mmHg) to mimic 5000 meters high above sea level, was utilized.
