Objective Pulmonic carcinoma is the most common thoracic cancer,including small cell lung cancer (SCLC) and non-small cell lung cancer (NSCLC).The pathogenesis of pulmonic carcinoma is not clarified, it's early diagnosis and curative effect are poor. Dihydrodiol dehydrogenase(DDH),as one member of aldo-keto reductase (AKR )superfamily,is one kind of plasmolysis protein. DDH plays important role in pathogenesis of lung cancer.To elucidate the feasibility of DDH as tumor markers in the diagnosis, it's concentration in serum of patients have been tested. DDH and vascular endothelial growth factor (VEGF) are inspected in tumor tissue to explore it's relations to cancer. The restrain of lung cancer cell line A549 by DDH2 antisense oligonucleotide is be observed in order to explore the possibility of it's clinical implication.Method 74 serum samples of patients with non-small cell lung cancer are respectively collected in Xiangya hospital in one week and one month postoperatively or prioroperatively.The concentration in serum of DDH is tested by enzyme-linked immunosorbnent assay(ELISA). 42 resected tissue samples of non-small cell lung cance is examined by immunohistochemistry to observe expression of DDH and VEGF and it's relations to clinical character of lung cancer. Non-small cell lung cancer cell line A549 is incubated and transferred by DDH2 antisense oligonucleotide in vitro,and the alteration of Dihydrodiol dehydrogenase protein and DDH2 mRNA is tested respectively by western-blot and RT-PCR in A549.The growth of A549 is observed by MTT and cell adherent rate.The nude mice model is designed,and DDH2 antisense oligonucleotide is injected in tumor.
