Objective:Acute and chronic exclusive reaction has constantly been an obstacle whichpuzzled us since long time ago although the mechanism of exclusive reactions after organ transplantation has been clear. There are many obviously toxic and side effects when using immunosuppressant. Although some success has been achieved in the experimental research for gene treatment, it is difficult for applying in clinic because of complex gene project and poor effect of medicine. Therefore, a feasible method should be searched to protect the transplanted organ be free of exclusion and not destroy the immune system of recipient. So our research aimed for: (1) To explore a high effective and simple method of separation and purification for testicular Sertoli cells and investigate the mechanism of immune-privileged origin from sertoli cell; (2) To establish a model of orthotopic liver with homologous chimeric testicular Sertoli cells, explore their effect and significance on FasLmRNA expression in liver tissue and estimate quantity of testicular sertoli cells request for the high amount of FasLmRNA expression; (3) To increase the stability and easy operating of orthotopic liver transplantation model effectively in rat by improving "two-cuffmethod"; (4) To...
