Objects To observe the change of glucagon gene transcription,translation and hormone release in high-fat diet induced insulin resistancerats.Methods Male Wistar rats were fed with regular chow or high-fat dietfor eight weeks, and were detected plasm glucagon level and glucagoncontent in islet by RIA, messenger RNA level in islet was assessed byreal-time fluorescence quantity RT-PCR.Results 1. Compared with normal control (NC) rats, glucagon releaseincreased significantly by 1.3 - 1.44 fold, glucagon content and messengerRNA level in islet increased by 1.3 fold and 1.35 fold respectively in insulinresistance (IR) rats.2. The insulin and glucagons ratio (I/G) in islet of IR rats increased significantly vs. NC rats, while the I/G ratio of diabetic IR rats decreased significantly vs. NC rats.Conclusions 1. Compared with NC rats, insulin and glucagon content in islet of IR rats increased significantly, which suggest that insulin couldn't inhibit glucagon biosynthesis in islet, and α-cell resistant to insulin that...
