Objectives:To culture and amplify mesenchymal stem cells from umbilical cord blood, and study the possibility of differentiating them into hepatocyte-like and islet-like cells in vitro. To observe the function of islet-like cells under the renal capsule of type 1 diabetic mice.Methods:1. Nucleated cells were separated by Hespan, and cultured with Mesencult? medium (containing 10% growth supplement).2. Expression of cell surface antigens and cell cycle were detected by flow cytometry.3. Cells were cultured in an adipogenic medium.The presence of adipocytes was assessed by the cellular accumulation of neutral lipid vacuoles that stained with Oil Red O.4. Cells were cultured in an osteogenic medium. The onset of osteoblasts was evaluated by the expression of alkaline phosphatase and calcium accumulation(von Kossa staining).5.
