Objective:To study the effect of cryopreservation on the growth of the human umbilical cord blood mesenchymal stem cells (UCBMSCs), and the potention of differentiation of UCBMSCs which were frozen and thawed and induced by Hepatocyte growth factor (HGF) and Fibroblast growth factor-4 (FGF-4) in vitro. The study also investigated whether the differentiated hepatocyte-like cells can induce UCBMSCs into hepatocyte-like cells.Method:1. The sixth-passage UCBMSCs were preserved by the cryoprotectant which consists of 10% dimethyl sulfoxide (DMSO) in liquid nitrogen. After 6 months, cells were thawed. Living cells were counted by Trypan Blue Dye Exclusion Method and observed by the inverted microscope.2.
