Experimental Study On Effect Of Lingqi Capsule On Anti-Tumor And It's Mechanism

Objective: To study the anti-tumor effect of Ling-Qi Capsules (LQC) and further explore the mechanism of action..Methods:1. Experiment in vitro: To observe the effect of the herb serum containing LQC on tumor cell morphology changes and to research its effect on the anti-tumor and/or inducing the apoptosis of K₅₆₂ leukemia cells with the methods such as MTT, AnnexinV/PI dual fluorescent staining, agarose gel electrophoresis, invert microscope and Wright-Giemsa stain in serum pharmacology.2. Experiment in vivo: To detect the expression of angiogenic factor cox-2 and bFGF in tumor-bearing mice with S-P immunohistochemical methods and to detect the LQC's inhibitory action to tumor angiogenesis by the content of iNOS in serum with colorimetric method and to study the inhibitory action to tumor by observation of tumor tissue pathological changes with optical microscopy and transmission electron microscope.Results:1. The herb serum containing LQC has evident inhibitory action on growth, proliferation and clone formation of human erythorleukemia cell line K₅₆₂ strain and this inhibitory action shows the dose-time dependent in 24-72h.2. The agarose gel electrophoresis shows typical apoptotic "ladders" when the herb serum containing LQC effect on human erythorleukemia cell line K₅₆₂ strain 72 hours.3. LQC has the anti-tumor effect to H₂₂ tumor-bearing mice,the inhibitory rate of high, middle and low dosage group is 35.74%,33.05%,25.09% and it show evident synergy and attenuation effect when using with Complex Prescription Cyclophosphamide.4. LQC can inhibit the tumor angiogenesis by down regulation the expression of angiogenic factor cox-2 and bFGF and degrading the contect of iNOS in serum of tumor-bearing mice.Conclusions:1. LQC has evident inhibitory action on growth and proliferation of K₅₆₂ strain and also can inhibite the proliferation of tumor cells effectively.2. LQC has the significant effect of inducing the tumor cells apoptosis.3. LQC shows its anti-tumor effect by influencing the expression of angiogenesis correlation factor and inhibiting the tumor angiogenesis.
