In these research, the recombinant adenovirus vector containing BMP-4 gene was prepared; the primary culture of human degenerative NP cells in monolayer was successful by an enzyme assimilation method; the results demonstrated innovatively that the BMP-4 gene can be expressed in human degenerative NP cells continuously and abundantly, and increase the production of intervertebral disc matrix (proteoglycans and typeâ…¡collagen ). It would break a new path for the gene therapy of intervertebral disc degeneration; the reseasch firstly analyses quantitatively the changes on the proteoglycans and typeâ…¡collagen gene in the development of degeneration of intervertebral disc after using recombinant adenovirus vector containing BMP-4 gene, in a animal model of intervertebral disc degeneration, which brought forward new considerations on the pathogenesis of intervertebral disc degeneration.
