ObjectiveUsing the important drugs from Loniceraeand Forsythiae Powder,including Lonicera japonica, forsythia suspense, great burdock acheneand their compatibility. To observe the effect of those extracts how toinhibite influenza A virus FM1 in vivo, including compatibility relation-ship, time-effect relationship, dose-effect relationship. Approach theimmunifaction mechanism of action to influenza viral infection. In orderto provide theory and experiment evidence so we can use traditional Chinesemedicine to prevent and cure influenza, and can provide theory and experimentevidence to prevent and cure flu.MethodUsing thin layer chromatography todetect the effective fractionsto anti-influenza virus in Lonicera japonica, forsythia suspense, greatburdock achene and their compatibility, and using HPLC to determinateeffective fractions quantitative of those medicine.Using influenza A virus FM1 to infect mice to induct pneumoniamodel, to study the compatibility relationship, time-effect relationship,dose-effect relationship of the important drugs from Lonicerae andForsythiae Powder. Comparing with the normal control,Ⅳcontrol group andvirazole group. Reviewing normal condition, lung exponent, lung constitutionpathological changes andⅣmRNA and IFN-γmRNA expression in lungconstitution, so we can approach the effect to restrain virus replicationand product IFN-γ. Using ELISA to detect IL-2 and IL-12 in lungconstitution suspension. Using immunity histochemical method to detectIFN-γand IL-4 proteinum express, to study and research the cell immunitymechanism to viral infection of above-mentioned medicine and theeffect to T cell subpopulation group balance.Result1. The quality assurance of drug empirical study shows the importantdrugs of honicerae and Forsythiae Powder contain anti-virus composition,caffeotannic acid, phillyroside and arctiin, and using HPLC to do quantitative study, and proved the single flavormedicine contents werethe tallest.2. Ribavirin and the important drugs of Lonicerae and ForsythiaePowder can restrain influenza virus. Comparing with the model group,they can cut down lung exponent, improve pathomorphology.Using immunityhistochemical method and ELISA to detect Th1 and Th2, the resulesshowed thatⅥRNA copy decreased and IFN-γmRNA expressed increasing.②The compatibility relationship reseach:Normal condition: The normal control animal appeared normal, hereafter one by one in order is the ribavirin group, No. 1 drug group, No. 2drug group, No. 3 drug group, No. 4 drug group, No. 5 drug group, No. 6 drug group,No. 7 drug group, the appearance of the model group was the worst.Lung exponent: Comparing with the model group, all the others hadvisible difference (P<0.05). The each set of Chinese herbal medicine andthe ribavirin group all could cut down the lung index number.The external appearance of lung organizes: The normal control was good,the next in order is the ribavirin group, No. 1 drug group, the changesbetween the others were alike, the model group was the worst.Lung constitution pathological change: the normal control had nochanges, the ribavirin group was the slight, the No. 1 drug group wasworse than it, the model group was the worst.Using RT-PCR to examine the virus nucleic acid and IFN-γmRNA:comparing with the model group, each set of Chinese herbal medicine andthe ribavirin group could restraint virus copy and product IFN-γ, butthe model group was the opposite.②The time-effect relationship of NO. 1 drug reseach:After different days fromⅣinfection, comparing with the modelgroup, NO. 1 drug and ribavirin group had different changes at position,lung exponent, lung constitution, and IL-2, IL-12, IFN-γcontents in lungconstitution, they all could inhibitⅣduplicating. Comparing withthe model group, Chinese medicion could improve the lung affection,pathological change, and could control the secretion of Thl cell factorIL-2, IL-12 and IFN-γ, Th2 cell factor IL-4.③The dose-effect relationship of NO. 1 drug showed, comparing with themodel group, the different dose of NO. 1 drug and Ribavirin group, all thedetections had significant deviation (P<0.05). Adding dose couldn'tenhance curative effect.Conclusion:The extracts from the important drugs of Lonicerae and ForsythiaePowder, including lonicerae, forsythia suspense, bardanae and their compatibility, has better therapeutical effect to mouse those wereaffected by influenza A virus FM1 strain, the mechanism maybe thesereasons:1. Relieving immunologic injury and restrain the duplication ofinfluenza A virus in mouse lung, extenuation the coup injury to lungorganize.2. Raising anti-infection cell mediated immunity:Raising lympholeukocytesubset Th1 cytokine IL-2, IFN-γ, IL-12, inhibiting Th2 cytokine IL-4,educing immunological regulation function, elevating immune response,enhance anti-virus capability.So the important drugs of Lonicerae and Forsythiae Powder can be theChinese medicine to cure Influenza virus, and maybe become immunologicaladjuvant to raise the immune fuction of vaccinum.
