Geranium is a kind of wonted Chinese drug. It has the function of eliminating thewind-damp, clearing the channels, and arresting diarrhea. It can be used in the symptoms ofarthralgia due to wind-dampness, anaesthetization and muscular constriction, bones and musclesache, and diarrhea in TCM. Through the first-hand investigation and market research we can findthat, besides the species of pharmacopoeia, many other species of Geranium are used as HerbaGeranii in different areas, of which 56 species belong to Geranium growing in China, and 13species are used in the Northeast of China. Thus the species are used confused. The thesis studiesthe main Herba Geranii on sale and the mixed species of Geranium in the Northeast of China. It setsup a macroscopical, organic, cellular and molecular level authentication method and it comparesand analyzes the quality systematically. At the same time, the thesis deeply studies the chemicalconstituents and pharmacodynamics of Geranium sibiricum L. thus lays the foundation for furtherdeveloping new medical plants resources.The first part: The authentication research of Herba Geranii. Adopt X-ray diffraction methodto authenticate different species of medical materials for the first time, create XRD recognitionmode, and analyze the degree of similarity of spectrograms; adopt IR spectrometry to finger printauthenticate and analyze the degree of similarity for the first time; adopt RAPD molecule labelingtechnique to compare and study different species Geranium, seeing about the genetic diversity andgenetic relationship for the first time, the method is accurate and reliable therefore create RAPDmolecule labeling technique for the medical material. Adopt routine powder flaking method tocompare the powder characteristics of roots, stems, leaves of different species; adopt olefin flakingmethod to compare the histological structure in transverse section of roots, stems, leaves, and calyxof different species. Adopt micro-quantifying to authenticate different species for the first time,adopt acetic anhydride-oil of vitriol method to compare the palynology characteristics of differentspecies; adopt routine flaking combined with photomicrography method to compare the karyotypeof different species chromosomes and undergo statistical analysis. Adopt the method of single UVspectral line combined with to UASLG to compare and authenticate different species. The thesisadopts many methods to authenticate Herba Geranii, change the limitation and inaccuracy of singlemethod. The thesis lay the foundation for the species differentiation and radicel authentication ofthe Geranium.The second part: Quality research of the Herba Geranii. Adopt UV spectrophotometry toanalyze the total flavone content of different Geranium species; adopt total phenols testing methodto analyze the total tannin content for the first time; adopt HPLC method to determine and comparethe gallic acid content.The results indicate the relationship of the components and the qualitydifference of different Geranium species.The third part: The chemical composition study of Geranium sibiricum L. Adopt solventextraction, silica gel and Sephadex LH-20 column chromatography methods to systematically separate and study the chemical composition of Geranium sibiricum L. Separate and identify 5compounds from the overground parts of Geranium sibiricum L. They are daucosterol, Kaempferol,Kaempferol-7-O-rhamnoside, Kaemferitrin, and Gallic acid.The fourth part: The pharmacodynamics study of Geranium sibiricum L. According to theanti diarrhea function of Geranium wilfordii Maxim, adopt the method of rats diarrhea by senna,apply the small intestinal movement experiments with charcoal powder in healthy mice,neostigmine—treated mice to compare the pharmacological action between Geranium sibiricum Land Geranium wilfordii Maxim. The results indicate that the Geranium sibiricum L extractingsolution can inhibit the rats diarrhea by senna, showing the anti diarrhea function; and the Geraniumsibiricum L extracting solution can inhibit the movement of the normal small intestinal movementand sthenic small intestinal movement by neostigmine, showing that Geranium sibiricum L caninhibit the movement of normal and hyperfumction stomach intestine.The fifth part: The quality standard research of Herba Geranii. Create more exhaustivequality standard for 3 species recorded in the 05 pharmacopoeia for the first time; create the totalflavone, total tannin, and gallic acid assaying methods for Geranium, undergo the microdifferentiation, TLC differentiation, and determine moisture, ash content, and extract.The thesis synthetically apply TCM Authentication, analysis of pharmaceutical dosage,pharmacology of TCM, statistics, computer technology and many modern research methods. Itcreate many differentiate modes and quality evaluation for many species of Herba Geranii, andsystematically study the chemical composition and pharmacodynamics of Geranium sibiricum L.The experimental technique is feasible and the results are accurate. It offers new clews andsignificant quests for systematically developing Herba Geranii.
