Objective:To observe the effect of scalp penetration acupuncture on Baihui Through Taiyang on the changes of 5-HT formation, metabolism and transport within hypothalamus of exercise fatigue rats, and discuss the mechanism and location of anti-exercise fatigue from the level of molecular biology. The main purpose is to take the advantage of acupuncture and refer objective experimental evidence on preventing exercise fatigue.Methods:Create the rat models of exercise fatigue by 4-week incremental swimming exercise, and randomly divide the rats into 4 groups:blank group, model group, Scalp-acupuncture group and traditional acupuncture group. Take the method of preventive acupuncture and record the exhausting swimming time at the 4th weekend. Draw material after exhausting swimming, take the methods of colorimetry, fluorospectrophotometer, high efficiency liquid chromatography and semi-quantitative RT-PCR, light microscope, electron microscope. And detect the changes of swimming time, LD, CK, BUN, Hb, BCAA, f-Trp,5-HT,5-HIAA, TPH, and the expression of MAO-A mRNA. The broad structure and ultrastructure of the hypothalamus.1,Scalp penetration acupuncture on Baihui through Taiyang can reduce the hypothalamus morphology of pathological changes and significantly increase the exhausting swimming time of big rats. In which the treatment group and model group were significantly different (P<0.01), treatment group, compared with traditional acupuncture group had significant difference(P<0.01).2,Scalp penetration acupuncture on Baihui through Taiyang can increase LD, CK levels in the treatment group compared with the other groups were highly significant differences (P<0.01); scalp penetration acupuncture on Baihui through Taiyang can reduce the BUN levels in the treatment group compared with the model highly significant difference (P<0.01), treatment group and control group, traditional acupuncture group showed no significant difference (P>0.05); scalp penetration acupuncture on Baihui through Taiyang can increase Hb levels in the treatment group and model group, the traditional acupuncture were compared with the very significant difference (P<0.01). treatment group and control group showed no significant difference (P>0.05);3,Scalp penetration acupuncture on Baihui through Taiyang can decrease the f-TRP within hypothalamus of exercise fatigue rats, the treatment group compared with the other groups are highly significant difference (P<0.01); Scalp penetration acupuncture on Baihui through Taiyang in rats after exercise training can increase serum BCAA of the treatment group and model group, compared to traditional acupuncture group were significantly different (P<0.01), the treatment group compared with the control group with significant difference (P<0.05); and inhibit f-TRP/BCAA levels, treatment group and control group, model group were highly significant differences (P<0.01), treatment group, compared with traditional acupuncture group had significant difference (P<0.05).4,Scalp penetration acupuncture on Baihui through Taiyang can reduce the producing of 5-HT, the treatment group compared with the other groups were highly significant differences (P<0.01); scalp penetration acupuncture on Baihui through Taiyang can increase 5-HT conversion rate, the treatment group and control group, compared to the model group were significantly different (P<0.01), treatment group, compared with traditional acupuncture group had significant difference (P<0.05).5,Scalp penetration acupuncture on Baihui through Taiyang can reduce TPHmRNA and deduce MAO-AmRNA expression in hypothalamus of rats, the treatment group compared with the other groups were highly significant differences (P<0.01).Conclusion:1,Scalp penetration acupuncture on Baihui through Taiyang is an effective way with multi-targets in exercise fatigue prevention.2,Scalp penetration acupuncture on Baihui through Taiyang can reduce fatigue hypothalamus Pathological changes in the organization, the organizational structure of the hypothalamus has a protective effect.3,Scalp penetration acupuncture on Baihui through Taiyang can significantly extend big rats'exhausting swimming time, improve exercise capacity in rats improve the metabolism of urea nitrogen and reduce muscle glycogen consumption to make homeostasis, and to delay the exercise fatigue.4,Scalp penetration acupuncture on Baihui through Taiyang can reduce free tryptophan and increase BCAA within serum, inhibit the expression of TPHmRNA and enhance the expression of MAO-AmRNA to inhibit the producing of 5-HT and accelerate the catabolism of 5-HT, which may be one of the mechanisms of preventing exercise fatigue.
