| ObjectiveA clinical research to explore the relationship between ROS and PMOP through observing the change of BMD,serum ROS in PMOP and Non-PMOP. An animal's experiment to observe the change of BW, BMD and ROS in kidney in the Ovariectomized rats. At the same time, applying Gu kang to affect these indicators, to investigate ROS's variation in PMOP and Gu kang's function of inhibiting ROS production.MethodThe clinical research:20 PMOP and 20 Non-PMOP patients (2010.2~2011.2) were divided into OP group and Non-OP group. We measured and observed the change of BMD, serum SOD, MDA and TAOC. In addition, We applied SPSS 16.0 software to analyze their correlations and significant differences in two groups.The animal's experiment:48 female 6-month-old SD rats were randomly divided into 4 groups of 12 each:the blank-control group (Hereinafter referred to as blank group, model(ovariectomized) group, observation (Gu kang)group and control (Western medicine)group, all of rats of the later 3 groups were ovariectomized. After the model of PMOP was established,4 groups were fed by gavage for 12 weeks. Then BW, BMD in femur, SOD, MDA and TAOC in kidney were measured.ResultsThe clinical research:1.BMD of OP group were lower than Non-OP group, it had a significant statistical difference (P=0.000<0.01). Serum MDA in OP group were higer than Non-OP group, it had a significant statistical difference (P=0.000< 0.01).However, serum SOD and TAOC in OP group were lower than Non-OP group, it had a significant statistical difference (P=0.000,005<0.01) 2. The result of correlative analysis between age, menopause time-limit and BMD:the correlation coefficient r=-0.368(P=0.020<0.05) and -0.360(P= 0.022<0.05), the scatter diagram showed that BMD had a tendency of decreasing along with the increasing of age and menopause time-limit, which indicated significant negative correlation between them.3.The result of correlative analysis between BMI and SOD, TAOC:the correlation coefficient r=0.403 and 0.479 (P=0.010,0.002<0.01), the scatter diagram showed that serum SOD and TAOC had a tendency of increasing along with the increasing of BMI, which indicated significant positive correlation between them.4. The result of correlative analysis between BMD and MDA:the correlation coefficient r=-0.892 (P=0.000<0.01), the scatter diagram showed that serum MDA had a tendency of decreasing along with the increasing of BMD, which indicated significant negative correlation between them. The result of correlative analysis between BMD and SOD, TAOC:the correlation coefficient r=0.501(P= 0.001<0.01) and 0.344 (P=0.030<0.05), the scatter diagram showed that serum SOD and TAOC had a tendency of increasing along with the increasing of BMD, which indicated significant positive correlation between them.5. The result of correlative analysis between MDA and SOD, TAOC:the correlation coefficient r=-0.589(P=0.000<0.01) and -0.397(P=0.011<0.05), the scatter diagram showed that serum MDA had a tendency of decreasing along with the increasing of serum SOD, TAOC, which indicated significant negative correlation between them.6. The result of correlative analysis between SOD and TAOC:the correlation coefficient r=0.393 (P=0.012<0.05), the scatter diagram showed that serum SOD had a tendency of increasing along with the increasing of serum TAOC, which indicated significant positive correlation between them.The animal's experimentFirst, Only forty SD rats were counted at the end of the experiment.Second, The BW of all rats had no significant statistical difference (P>0.05) in before modeling and administration. But after 12-week administration, the BW of all rats was increased, comparision with before administration, it had significant statistical difference(P<0.01);the BW of model group was higer than blank group and control group, there were significant statistical differences(P<0.01), but it had no significant statistical difference (P>0.05) in model group and observation group.Third, After 12-week administration, whole section of the left femur BMC and BMD in blank group and model group had significant statistical difference(P<0.01);Comparision with whole section of the left femur BMD in model group, observation group and control group had significant statistical difference(P<0.05);But it had no significant statistical difference (P>0.05) in observation group and control group. Whole section of the left femur BMD in model group was lowest of 4 groups.Finally, After 12-week administration, MDA in kidney was obviously increased in model group, Comparision with blank group, it had a significant statistical difference (P=0.036<0.05); SOD, TAOC in kidney had a tendency of decreasing in model group, Comparision with blank group, it has no significant statistical difference (P=0.538,0.164>0.05). Comparision with model group, SOD in kidney were obviously increased in observation group and control group, it had a significant statistical difference (P=0.007<0.01, P=0.033<0.05);TAOC in kidney had a tendency of increasing and MDA in kidney had a tendency of decreasing in observation group and control group, comparision with model group, it has no significant statistical difference (P=0.123,0.104,0.761,0.429>0.05). MDA, SOD and TAOC in kidney had no significant statistical difference in observation group and control group(P=0.639,0.483,0.964>0.05).ConclusionClinical research conclusion:1.High level MDA and lower lever SOD, TAOC in PMOP illustrated oxidative damage had occurred in PMOP. The balance of oxidation and antioxidant had been broken in the body.2. BMD were significant decreased in PMOP, and there were significant negative correlation between BMD and age, menopause time-limit.3.There were significant negative correlation between serum MDA and SOD, TAOC, and significant positive correlation between serum SOD and TAOC in PMOP. Which illustrated MDA and SOD, TAOC are closely related with the disease and influence each other. There were significant negative correlation between serum MDA and BMD, and significant positive correlation between serum SOD, TAOC and BMD, BMI in PMOP. Which illustrated abnormal secretion between MDA and SOD, TAOC will lead to lipid metabolism disorder and decreased BMD in PMOP. Imbalance between MDA, SOD and TAOC probably have a close relation with PMOP.4. These oxidation and antioxidant indicators are secondary indicators, which can be used to evaluate osteoporosis severity.Animal's experiment conclusion1. After ovariectomized, BMD decreased, BW increased, at the same time, there were high lever MDA and lower lever SOD, TAOC in kidney, which indicated lower lever E2 had led to osteoporosis, imbalance in lipid metabolism and oxidative damage in the rats.2. Two medicine had similar effect, they can inhibit BW gain and increase BMD in ovariotomized rats.3. Two medicine can obviously increase SOD content, mildly increase TAOC content and decrease MDA content in kidney, which illustrated they can prevent oxidative damage.4. Gu kang's mechanism of antioxidant damage is related to the reasonable drug formulation and have estrogen-like and inhibit ROS production substances in the medicine. |
