Herb Lamiophlomis is recorded in 'Pharmacopoeia of the People's Republic of China'(2010 edition). It is the dried aerial part of Lamiophlomis rotata (Benth.) Kudo (Fam. Labiata). sweet, bitter in flavour, neutral, acting on the liver channel, and good at promoting blood flow to stop bleeding, eliminating wind, relieving pain and curing impetigo, usually being used to treat traumatic injuries and bleeding, rheumatic arthralgia and impetigo.ObjectiveThis study is a part of'Development and Research of Herba Lamiophlomis'project which is under the State Ministry of Science and Technology 11th Five-Year Plan Support (No. 2006BAI06A18-19). Considering the research status and problems of Herba Lamiophlomis, our study aimed at active fraction, quality standards, method of original processing, effect of prepair processing, mechanisms, pharmacokinetics, expecting to establish a more scientific quality standards to conduct original processing and prepair processing, and to discover the mechanisms to provide the basis and idea for further study.MethodsThe methods used in the research of the original processing and the mechanism of Herba Lamiophlomis are philological, pharmacological and chemical means.1. Systemically arrange the documents about application history and follow up the modern report timely by philological means in order to recognize the research status and problems of Herba Lamiophlomis.2. Basing on the new quality standards, research the methods of original processing, prepair processing, and the effect of prepair processing of Herba Lamiophlomis.3. Separate the active ingredients which concern with the effect of relieving pain directly and have strong specificity by chemical means, and establish a new quality standards with the new determination indicators.4. Research the mechanisms of relieving pain and anti-inflammatory of Herba Lamiophlomis by pharmacological means.5. Using the determination indicators of the new quality standards, process the pharmacokinetics studies.Results1. The results showed that dry temperature, storage humidity made a great impact the content of flavonoids and iridoid of Herba Lamiophlomis, and wine-process could rise the transfer rate of flavonoids and iridoid of Herba Lamiophlomis.2. The results showed that the active fraction of relieving pain of Herba Lamiophlomis was n-butanol fraction. Shanzhiside methyl ester and 8-O-acetylshanzhiside methyl ester were isolated and identified from the active fraction. The new quality standards with the two chemical composition as determination indicators can control the quality of Herba Lamiophlomis better.3. The results showed that Herba Lamiophlomis could control the leukocyte count in joint capsule synoviocytes of rats with adjuvant arthritis, inhibit releasing histamine of rats with arthritis induced by carrageenan, control the level of inflammatory mediators of TNFα,IL-1 in blood serum, and rise the level ofβ-endorphin in hypothalamus of model animals.4. The body process of shanzhiside methyl ester meet the three compartment model afer intravenous administration, the main absorption in rats showed a linear dynamic process, and meet the two-compartment model after intragastric administration the main absorption in rats showed a linear dynamic process. The body process of 8-O-acetylshanzhiside methyl ester meet the three compartment model afer intravenous administration, the main absorption in rats showed a nonlinear dynamic process, and meet the two-compartment model after intragastric administration the main absorption in rats showed a nonlinear dynamic process.ConclusionsDry temperature, storage humidity can make a great impact the content of flavonoids and iridoid of Herba Lamiophlomis, and wine-process can rise the transfer rate of flavonoids and iridoid of Herba Lamiophlomis. Shanzhiside methyl ester and 8-O-acetylshanzhiside methyl ester were the active composition of relieving pain of Herba Lamiophlomis. The quality standards with the two chemical composition as determination indicators can control the quality of Herba Lamiophlomis better. The mechanisms of relieving pain and anti-inflammatory of Herba Lamiophlomis probably concern with controling the leukocyte count in joint capsule synoviocytes, inhibiting releasing histamine, controling the level of inflammatory mediators of TNFα,IL-1 in blood serum, and rising the level ofβ-endorphin in hypothalamus. The metabolic process of shanzhiside methyl ester and 8-O-acetylshanzhiside methyl ester is complicated, need further research.Innovations1. Researched the original processing and prepair processing of Herba Lamiophlomis, provided scientific basis for pre-process of Herba Lamiophlomis.2.Shanzhiside methyl ester and 8-O-acetylshanzhiside methyl ester which concern with the effect of relieving pain directly and have strong specificity were isolated and identified from Herba Lamiophlomis. Established a new quality standards of Herba Lamiophlomis with the new determination indicators.3. Made an exploratory study of mechanisms and pharmacokinetics of Herba Lamiophlomis, provided reference for further elucidating the mechanisms of Herba Lamiophlomis.
