Objective:under the direct of Chinese medicine diagnosis and treatment theory, and the guidance of long-term clinical practice, the subject observes the effects of ShenQi Compound Recipe on Nuclear FactorΚB(NF-ΚB) and Peroxisome Pro-liferator-activated Receptors gamma(PPARγ) in Kidney of GK Rats, to further explore the mechanisms of ShenQi Compound Recipe in treating Diabetic Mellitus(DM) and Diabetic Nephropathy(DN), so. theoretical and experimental basis for he rational use of ShenQi Compound Recipe and the development of new drugs can be provided.Methords:45 GK (Goto-Kakizaki) rats, SPF(specific pathogen free)grade, male,16 weeks old, After being adaptively feeded for 1 week, whose RBG≥11.1mmol/L were randamly divided into western medicine group, Shenqi Compound Recipe high-dose group, Shenqi Compound Recipe low-dose group, model group; and 10 SD rats (SPF grade) were set as the control group with normal diet for 8 weeks, while the other groups were fed with high fat diet. The western medicine group were fed with Ramipril suspension by lmg/(kg·d); Shenqi Compound Recipe high-dose group were fed with the ShenQi Compound Recipe suspension by 2.88g/(kg·d); Shenqi Compound Recipe low-dose group were fed with the ShenQi Compound Recipe suspension by 0.72g/(kg·d); Model group and control group were fed with sterile water by 5ml/(kg·d). During the period of experiment, general state, weight, quantities of eating and drinking, and random blood glucose of all rats were monitored. After 8 weeks the Tail capillary fasting blood glucose were tested by Glucose oxidase way, the cholesterol and triglycerides were measured by Enzyme-coupled assay way, the Creatinine, and Urea nitrogen were measured by oxidization way, IL-6 was measured by Enzyme-linked immunosorbent assay(ELISA) way. The right kidneys were taken, part of every right kidney for Morphological observation by HE staining, and the average integrated optical density changes of NF-ΚB and PPARγBy immunohistochemical staining (SABC) way; Part fixed for electron microscopy sections to observe ultrastructural changes. The rest were cooled by liquid nitrogen immediately for RT-PCR detection of NF-ΚBmRNA and PPARγmRNA.Results: Shenqi Compound Recipe high-dose group GK rats are Significantly better than the model group in general state, weight gaining, food and water intaking; the blood glucose, lipid, IL-6 Lower than the model group, there is significant difference between the two groups (P<0.05,&P<0.01); Pathological changes significantly reduced than the model group. expressions of NF-κB in Chinese medicine high-dose group decreased, while Expression of PPARγincreased, there is significant difference between the two groups (P<0.05,&P<0.01)Conclusions:Shenqi Compound Recipe high-dose can lower the blood glucose, blood lipids, IL-6 levels of GK rats, inhibit activities of NF-κB, while increase the expression of PPARγ, Delay the renal pathological changes of GK rats, which may be one of the mechanisms of preventing and treating DM and DN.
