| Objective: FengShi Ning(FSN) capsule is used as a therapeutic option for animals with experimental RA. The aim of this study was to investigate the effect and mechanism of FSN on the AA. Methods: Take the adjunvant arthritis (AA) rats as animal model for RA, which were treated with FSN and tripterygium wilfordii polyglycosidum(TWP) respectively. And the expression of ICAM-1, uPA and MMP-3 was observed by immunohistochemical examination. Results: (1) FSN could inhibit secondary joints swelling and multi-arthritis evidently, improve the whole condition of rats; at the same time, it can also lighten the synovial inflammation and hyperplasia of lesion joints distinctly, and prevent the joint cartilage and bone from destruction; the collective efficiency of FSN is better than TWP. (2) Expression of ICAM-1 was observed on synovium and chondrocyte. FSN could decrease the expression of ICAM-1, there were significant difference between FSN group and model group( P<0.01) (3) FSN could depress abnormal hyper-expressicn of uPA in synovial tissues and chondrocyte (P<0. 01) (4) FSN can lower the expression of MMP-3 in synovial tissue and chondrocyte , thus inhibit the damage of cartilage and bone. (5) The above effect of FSN are in proportion to its dosage. Conclusions: The data suggest that FSN has inhibitory effects on symptom and condition of experimental RA, which is better than TWP as a whole. The anti-inflammatory effect is related to its adjustment effect on the expression of ICAM-1. The lower expression of uPA and MMP-3 by FSN in joint synovial tissue and chondrocyte, is probably the basis of controlling immflammation and proliferation of synovium and the destruction of joint cartilages and bones so as to retard the disease progressing. |
