Blood Side Of Qilong Tune The Treatment Of Immune Thrombocytopenic Purpura And Mechanisms

1 Objective Immune thrombocytopenic purpura (ITP) is a common hemorrhagic disease in clinic.Because its morbid reason and mechanism are not clear until today, its clinic curativeeffect is limited . We planed to establish the animal model with ITP integratedsyndrome in TCM without any intervenient methods,at the same time,we studied thecurative effect of QiLongTiaoXue Recipe and explored its effective mechanism . Wealso tried to test and verify the feasibility of establishing animal model with ITPwhich can integrate disease and syndrome in TCM on combining macroscopic method withmicrocosmic method,and provide experiment basis for applicating of QiLongTiaoXueRecipe in clinic and exploiting of new medicine.2 Methods The animal model of ITP was established by using the method of immunization .According to the counts of blood platelet, we divided 24 BALB/C mice into 2 differentgroups randomly :the normal and the model group . APS which dilite concentration was1:4 was injected into abdominal cavity of the model mouse at a dose of 100μl/20gevery other day . We detected the counts of blood platelet, the counts and classifyof the megakaryocyte in bone marrow, the change of pathology of thymus and spleenafter 2 weeks in order to judge whether the model was successful or not. To judgethe feasibility of establishing animal model with ITP which can integrate diseaseand syndrome in TCM, we observed dynamically the syndrome in TCM of model mice,suchas:the spirit condition ;action ;defecate;urinate; hair;reation;gesture;ecchymosisand petechia on the skin;capacity for eating and drinking ; temperature;weight ;swimming time ; wither and blood stasis or hemorrhage in organs or tissues. After the animal model with ITP integrated syndrome in TCM was verifiedsuccessful ,we divided 84 BALB/C mice into 7 different groups randomly according tothe counts of blood platelet, those are :the normal,the model,the Xuekang oralliquid,the prednisone,the big,medium and small dosage of QiLongTiaoXue Recipe groups .One week after establishing the model ,we gave orally physiological salt solutionand every treated medicine daily for additional 1 week. We evaluated the curativeeffect of QiLongTiaoXue Recipe according to detecting the counts of blood platelet,the counts and classify of the megakaryocyte in bone marrow,the blood coagulate time,the change of pathology of immunological organs , the degree of hemorrhage and thechange of syndrome in TCM. After the curative effect of QiLongTiaoXue Recipe had been defined,we establishedthe model according to the methods above-mentioned and took the big dosage ofQiLongTiaoXue Recipe as the treated medicine.We detected the changes of theglycoprotein on the membrane of the blood platelet and the T-cell subgroup with themethod of flow cytometry,and examined the levels of the IL-2,6,8 with the methodof radiate-immunity. We also detected the apoptosis rate of the thymus and spleenlymphocytes and expression of the Fas and Bcl-2 gene with the TUNEL method and theimmunohistochemistry method .4 芪龙调血方治疗免疫性血小板减少性紫癜疗效与机制研究3 Results The results showed: ①Compared with the normal group , the counts of blood plateletof the model mice decreased obviously, the counts of megakaryocyte in bone marrowof the model group mice increased,but the megakayocyte which can produce bloodplatelet decreased.The model mice had the clinical feature of "Qi-Deficiency andBlood Stasis " . The results of experiment were in accordance with clinicalmanifestation. ② The counts of blood platelet recovered after treated withQiLongTiaoXue Recipe ,compared with the model group ,it had marked statisticalsignificance (P < 0.05), the big dosage of QiLongTiaoXue Recipe recovered mostobviously.The hyperplasia of the megakaryocyte in bone marrow which had maturedisturbance was restrained in the big and medium dosage of QiLongTiaoXue Recipe groupsand the megakaryocyte whic...
