| Objective: To analyze the expression of loss of hererozygosity(LOH) and microsatillite instability (MSI) of tumor DNA and plasma DNA, and to investgate their relations. To explore the value on the microsatillite change of plasma DNA for the early diagnosis, screening and follow-up.Methods: 43 cases of laryngeal cancer treated surgically were analyzed . Preoperative and postoperative blood(10ml), tumor tissue, para-tumor tissue were collected. The lymphocyte and plasma was separated. The DNA was extracted respectively. Three loci of D9S162, D9S1782, D17S1665 with high occurrence were selected , the primer was projected and composed. The electrophoresis and DNA sequence measurement were performed with ID Image Analysis Software and ABI Prism 3100 Genetic Analyzer after PCR.Results: The incidence of LOH, MSI, AI of tumor, preoperative plasma, postoperative plasma for D9S162 was 51.85%, 22.22%, 74.07%, 44.44%, 18.52%, 62.96%, 33.33%, 14.81%, 48.15%; And for D9S1782: 43.48%, 13.04%, 56.52%, 39.13%, 13.04%, 52.17%, 34.78%, 13.04%, 47.83%; D17S1665: 51.43%, 17.14%, 68.57%, 42.86%, 14.29%, 57.14%,... |
