Rat Model Of Adenine Caused Spermatogenic Cell Damage. Bushenshengjing Pill Intervention Effect And Its Mechanism Research

ObjectiveWe summarize the realization in TCM and status quo in modern medicine of male infertility, and feed the model animals of deficient kidney yang caused by amidopurine with Bushen Shengjing Pill (BS) so as to probe the mechanism of action and build the biological theory of kidney dominating the reproduction and produce the experimental base for pharmacology.MethodsSubgrouping and administration: divide the 96 SD rats of 3 month in age into normal group, model group, large dose of BS and small dose of BS groups at random, 24 rats in each group. The rats in the normal group are fed with 5% sodium carboxymethycellulose, 10ml/kg; the rats in the model group are fed with amidopurine fluid, 250mg/kg; the rats in the large and small dose group of BS are fed with 400mg/kg and 200mg/kg respectively, and then fed with midopurine 250mg/kg 40 minutes later. The whole experiment lasts 40 days. Take 8 rats in the normal and model group, feed them with drugs on the 20th, 30th and 40thday, calm them with 10% chloralization after 1 hour of last feeding, take the abstraction serum from the abdominal aorta and wait for the test of biochemistry, enzymology and apoptosis. Test the endocrine in the serum, the content of SOD and MDA in the testicle tissue and the activity of the NOS and the content of NO in the testicle. Watch the change of histomorphology in the light microscope; test the apoptosis condition with TUNEL method.ResultsOn the 30th day, there is obvious difference among the different model groups in sperm activity ratio and there is obvious difference between model group and the other three groups in Sperm count, LH value, T value, activity of SOD, content of MDA, activity of NOS, and content of NO. But there is no difference in FSH value. Comparing with the normal group, the contorted seminiferous tubules of the rat's testicle in the model group goes slightly shrink, the lumina endepidermis goes thinner, the sperm and androgonial cell become less, there is interstitial edema, and interstitial cell becomes vacuolar degeneration.There is obvious difference change in spermatogenic cell apoptosis. It can be concluded that the model on the 20th day is made in success.On the 40th day between the model and normal groups, there is obvious difference in sperm activity ratio, Sperm activity,Sperm count, LH value, T value, activity of SOD, content...
