"triple Therapy" Rabbit Osteoarthritis Of The Knee Cartilage Matrix Metalloproteinases And Their Inhibitors

Objective: To study the pathogenesis of knee OA and the mechanism of the three united remedy (Chinese medicine fumigation, the massage, functional training). The project studied the mechanism of OA with the cell messenger factors and explain the basic thoery of the three united remedy for OA.Method: â‘ S.D rabbits were divided into 7 groups:normal group, the model group,the three united remedy treating group,the fumigating group,the functiong training group,the massaging group,the Votalin treating group. Rabbit knee OA high pressure in bone model was builded by means of ligating the right femoral vein to block lower extremity vein part recirculation of test side.All groups took 4 weeks treatments. â‘¡ Survey the ultrastructure changse of the each group knees. â‘¢ Survey the expression of MMP-1 and TIMP-1 by immunohistochemistry.â‘£ Survey the mRNA expression of MMP-1 ,-13 and TIMP-1 by RT-PCR.Results: â‘ The knees of model group were non-gloss,opaque,with loss and new growth,cartilage cells necrosis, arranged disorderly, and obvious anomaly. The group of the three united remedy can improve the model rabbit knee cartilage damage, and the effect was much better than the other groups. â‘¡The expression rates of MMP-1 and TIMP-1 :MMP-1 in the model rabbit knee cartilage increased, but TIMP-1 was negative. MMP-1 positive expression rates of control groups decreased ( P<0.01 ) . The group of the three united remedy decreased significantly ( P<0.05 ) than control groups. TIMP-1 expression rates of control groups were quite low, and no significant difference. â‘¢ The positive expression rates of MMP-1 and TIMP-1: the mRNA expression rates of MMP-1 ,-13 and TIMP-1 in knee cartilage of nomal groups were very low. The mRNA expression rates of MMP-1 ,-13 and TIMP-1 in knee cartilage of model group significantly increased than nomal group( P<0.01 ) . The expression of TIMP-1 was negative in model group.
