Tumor Necrosis Factor of human(TNF), which takes part in many physiological and pathological processes, is a kind of important cytokines. The effects of bioactivity is made by binding with specific membrane TNF receptors (TNFR) which are present on a wide variety of different cell types. Two types of TNFR are identified as type I receptor (TNFRI) and type II receptor (TNFRII). Soluble forms of TNFR were generated by proteolytic cleavage extracelluar portion to downmodulate membrane-bound receptors the of the two type receptors. As TNFR, there are two types of sTNFRs. In this study, using man type I souble TNFR(sTNFRI) which was cloned and expressed in insect cell Sf9, as targets , an innovative approach, nonradioactive receptor binding assays has been employed for screening sTNFR antagonist. Specially prepared cell-free systems have allowed the use of high concentrations of microbial metabolites in the reaction mitures with low percentage of false positive. More than 2000 microbial samples from different species of soil isolates and 50 kinds of Chinese herb samples have been tested. Using the bioassay, we got two interesting activity microbial strains and two herbs.
